Objective: To investigate the inhibitory effect of combination of severaldifferent cytokine genes on the H22tumor.Analyzing the best concentrate andcombination of the gene therapy on H22tumor.Methods:(1) Tumor model was established by inoculation of H22hepatocarcinoma cells at right hind thigh of BALB/c mice.(2) Tumor gene therapywas performed by in vivo gene transfection with the method of injection of nakedplasmid into tumor to express polypeptide IFN-β、TNF-α、CXCL-1、CH50.(3) Tumorgene therapy was performed by in vivo gene transfection with the method based onhydrodynamics to express polypeptide IFN-β.(4) Tumor weight was used to reflectthe the inhibitory effect of different polypeptides on tumor growth.(5)Combination of the best dose of different polypeptide to the tumor-bear mice, andevaluate the advantages of the combination therapy.Results:(1) The best dose of the pcDNA3.1-IFN-β plasmid is100μg/100μlwith the hydrodynamic tail vein delivery method.(2)The best dose of thepcDNA3.1-TNF-α plasmid is10μg/100μl with injection into into the tumormicroenvironment.(3)The best dose of the pcDNA3.1-CXCL-1plasmid is10μg/100μl with injection into into the tumor microenvironment.(4)Combining above threeplasmids, we can get stronger suppressive effect on tumor growth than any one ofthem.Conclusions: Combining a variety of antitumor cytokine’gene with differentways has a better antitumor effect than using one kind of cytokines at one time. |
