Objective: To investigate the curative effect and expression ofCaspase-3mRNA、SurvivnmRNA of xingding joint THP on bladder cancer cellT24transplantation tumor in nude mice. It can provide the theory basis forxingding as the clinical adjuvant application in bladder perfusion chemotherapy.Methods:48nude mice were randomly divided into8groups aftersubcutaneous injection of T24cell into the right legs, the control group; threexingding group, treated with xingding of high (150mg/kg), medium(100mg/kg) and low-dosage (50mg/kg) respectively; two pirarubicin (THP)group, treated with THP of routine dosage (15mg/kg) and low-dosage(7.5mg/kg) respectively; and two combined treatment groups, A treated withxingding of high-dosage+THP of low-dosage (7.5mg/kg) and B treated withxingding of high-dosage+THP of routine dosage(15mg/kg); They were givenNS or THP into the tumor once every3days, given different dose xingdinglavage once every day, starting on the14th day after tumor forming. Tumorvolumes were measured every3days for drawing the growth curve1. Animalswere executed on the21th day after treatment. The inhibition rate of tumor wascalculated,at the same time also the interaction of the two drugs calculated by jin ’ s formula:xingding and THP. Light microscopy was used to observe themorphological changes. RT-PCR was used to detect Survivin、caspase-3mRNAexpression. Results: The average volume of tumor were682.0±36.6mm3inxingding of low dosage group,562.4±32.3mm3in xingding of medium dosagegroup,496.0±15.6mm3in xingding of high dosage group,505.0±35.9mm3inTHP low dosage group,389.3±26.5mm3in THP routine dosage group,375.3±23.5mm3in A group,302.5±15.0mm3in B group. They were all lowerthan the control group’s average volume857.6±71.8mm3(P<0.05). The averageweight of tumor were1.08±0.25g in xingding of low dosage group,0.94±0.17gin xingding of medium dosage group,0.86±0.16g in xingding of high dosagegroup,0.78±0.15g in THP of low dosage group,0.61±0.13g in THP of routinedosage group,0.37±0.07g in A group,0.50±0.09g in B group. They were alllower than the control group’s average weight1.26±0.20g (P<0.05). accordingto the inhibition ratios of the tumor weight,A group’s drug interaction(q) was1.04, B group’s drug interaction(q) was1.11. showed that the two kinds ofmedicine have additive action. Local necroses were observed in the tumorspecimens by HE staining in xingding group, THP group and Xingdingcombined THP group. The Survivin mRNA expressed down-regulatedly whentreated by Xingding of high-dosage combined with THP of low-dosage and THPof routine-dosage respectively (P<0.05), the effect is better than THP. theCaspas-3mRNA expressed up-regulatedly after treated by Xingding ofhigh-dosage combined with THP of low-dosage and THP of routine-dosagerespectively (P<0.05), the effect is better than THP. Conclusions: Xingdinghave a inhibition on the growth of transplanted tumor,in combination with THPcan strengthen the inhibiting effect of xenograft proliferation, and the effect isbetter than use THP alone. the Survivin expression down-regulation and the Caspase-3gene expression up-regulation is possible primary molcularmechanism,its molcular mechanism is undefined,need further study.
