Purple sweet potato, convolvulaceae, was famous of rich in anthocyanins and flesh. It contained many kinds of components with biological activation, such as proteins, flavonoids, polysaccharides, which were active and diverse. So, purple sweet potato had good application prospects.(1) Firstly, the system components’pre-test were conducted, and measured the contents of major nutrition. The extraction way is:firstly, extract protein, secondly, extract flavonoids, thirdly, extract polysaccharides.(2) According to single factor experiments and central composite experiments, we got the optimum extracted conditions of proteins:the rate of liquid/material70mL/g, the ultrasonic power390W, the extraction time21min, the extraction temperature20℃, the extraction times1time, the extraction yield was3.97%.(3) According to single factor experiments and central composite experiments, we got the optimum extracted conditions of flavonoids:the concentration of ethanol85%, the rate of liquid/material51mL/g, the ultrasonic power410W, the extraction time20min, the extraction temperature33℃, the extraction times1time, the extraction yield was3.19%.AB-8macro porous resin was the best resin for purificating purple sweet potato flavonoids, the optimal conditions were:the original concentration0.9mg/mL, the liquid velocity2BV/h, wash with water to pH4.0, the desorption agent was70%ethanol, the liquid velocity1BV/h, the amount of desorption eluant3BV. The purity of flavonoids was72.3%after purification with AB-8resin.HPLC-MS and HPLC tests illustrated that FPSPL hydrolyzed sample contained Rutin, Quercetin, Kaempferol, Leteolin and Isorhamnetin.(4) According to single factor experiments and central composite experiments, we got the optimum extracted conditions of polysaccharides:the rate of liquid/material80mL/g, the ultrasonic power395W, the extraction time30min, the extraction temperature69℃, the extraction times1time, the extraction yield was3.66%.Compared decolorization effects with activated carbon and macro porous resin, OU-1resin was the best resin for decolorization. The optimum conditions:the pH4.0, room temperature.(5) The research of mice illustrated that, flavonoids of purple sweet potato leaves could decrease Liver Index, the activity of serum Alanine Aminotrasferase and Aspartate Aminotrasferase, the level of Malondialdehyde in liver homogenate; increase the concentration of serum Albumin and Total protein, the activity of Superoxide dismutase in liver homogenate to mice with chronic liver injury induced by carbon tetrachloride. Flavonoids of purple sweet potato leaves had remarkable protective effect on mice with chronic liver injury induced by carbon tetrachloride, at a dose level of50mg/kg-d.
