| American Ginseng refers to members in the family Araliaceae within the generaPanax and Eleutherococcus. In recent years, more and more research scholars in the worldaccepted it because of its special properties.Taking American ginseng as raw materials, studied the traditional extraction methodsof ginsenosides. In order to extract more ginsenosides, we used12times the amount ofethanol on the basis of original traditional extraction method, at the same time, removedimpurities such as pigments by macroreticular resin adsorption; Then used vacuumconcentration extract ginsenosides, After freeze drying, we determine the totalginsenosides by HPLC. Finally by the method of chemiluminescence, we studied theantioxidant activity of the total ginsenosides and ginsenoside Rb1.Used Alcohol-resin adsorption method to extract ginsenosides, explored a simple andhigh extraction method. Compared vacuum freeze drying with traditional drying methodFrom the appearance, odor, drying time, Amount at Stake of ginsenosides andreconstitution properties. We determined the advantages of the vacuum freeze dryingmethod in drying ginsenosides. The extraction of ginsenoside Rb1and Re was determinedby HPLC from the total ginsenosides, and compared the different contents betweenginsenoside Rb1and ginsenoside Re. Experiment using Agilent1100system, AgilentExtend-C18column (5μm,2504.6mm) and column temperature was35℃,wavelength Selection was203nm, the flow rate was1.0mL/min, nitrile and ultrapurewater were the moving phases, the ratio is7:3, sample size was20μL. Determinationresults show that the content of ginsenoside Rb1and Re are higher under the experimentalmethod of extraction and separation of monomer, the method is simple and effective interms of analysing ginsenosides. In order to test the antioxidant activity of American Ginseng. The provention effectof radical scavenging activity of superoxide anion (O2-·) and hydroxy radical (·OH) wereevaluated using chemiluminescence (CL). Using the CuSO4-Luminol-Vc-H2O2system toevaluate the hydroxy radical (·OH); the Pyrogallol-Luminol system to evaluate the radical scavenging activity of superoxide anion (O2-·). Results showed that the extracts of thetotal ginsenosides had strong ability of eliminating·OH and O2-·, the IC50were1.23mg/mL and0.048mg/mL, showed a strong antioxidant activity; Ginsenoside Rb1extracted from American Ginseng had less strong ability the ability of eliminating·OHand O2-·, the IC50were3.19mg/mL and2.20mg/mL, also showed a strong antioxidantactivity. Experimental results showed that with Vc as positive control, both the totalginsenosides and ginsenoside Rb1showed higher antioxidant activities than Vc, andPresented the concentration-response relationship. |
