The phenolic compounds, including phenolic acids, flavonoids and other compounds, existing in the natural plants widly, is a kind of biological and pharmacological activity compound. The high performance liquid chromatography-electrochemical detection (HPLC-ECD) is used in analysis of multi component samples widely. In this thesis, the new method of HPLC-ECD with liquid-liquid extraction and liquid-solid extraction is established to detect thirting phenolic compounds in Chinese traditional medicine and honey. The present studies hold a certain significance in broadening the application range of HPLC and in quality control of Chinese traditional medicine and honey. This thesis is consisted of four chapters. The author's main work is as follows four points:1. A new method of HPLC-ECD is established for the determination of chlorogenic acid and baicalin in Kanggan Jiedu granules. The experimental results indicate that the calibration curves of chlorogenic acid and baicalin show good linearity with their peak areas in the certain concentration ranges(r>0.9995). The average recoveries of chlorogenic acid and baicalin are 98.3% and 97.2%, and relative standard deviations are respectively 1.3% and 2.4%. The method is rapid, sensitive, accurate, which can provide a basis for quality control of Kanggan Jiedu granules.2. A new method of HPLC-ECD using liquid-liquid extraction is established for simultaneous separation and determination of three synthetic phenolic antioxidants (SPAs), including butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT), tertiary butyl hydroquinone (TBHQ) in food items. The results show that the calibration curves of BHA, BHT and TBHQ show good linearity with their peak areas in the certain concentration ranges (r>0.9997). The average recoveries of three SPAs ranged from 95.0-104.1% with RSD<2.3%, and the detection limit is 2.0×10-2μg·ml-1. the method can be used to test oil and food items products.3. A new method of HPLC-ECD is established for the determination of Gallic acid, caffeic acid, ferulic acid, quercetin and keampferol in rape honey. The experimental results indicate that the calibration curves of five compounds show good linearity with their peak areas in the certain concentration ranges (r≥0.9997). The average recoveries of all the five compounds ranged from 97.4% to 101.6% with RSD<2.1%. Compared with HPLC-DAD, HPLC-ECD is much more sensitive in determination of the phenolic compounds.4. On the study of rape honey from ten different regions in Shanxi, the rape honey fingerprint of a phenol-based antioxidant compounds is established by HPLC-ECD. The 17 common peaks and 5 phenolic compounds index peaks from 10 different samples are identified by the rape honey fingerprints. On the study of 12 kind of jujube honey from different provinces, the jujube honey fingerprint of phenolic compounds is established by HPLC-ECD. The 13 common peaks and 5 phenolic compounds index peaks from 12 different samples are identified by the jujube honey fingerprints. The research can provide reference for identification of Chinese jujube honey and Shanxi rape honey and distinguishing true from false honey.
