With the advent of the aging population, aging-related diseases occur, which receive more and more attention. The occurrence of aging-related kidney disease grow with each passing day. It is thought that klotho gene and the occurrence of aging is closely related, the reduction of its expression seriously affects the quality of life and life expectancy of older persons, and the performance of its impact on quality of life can be sexual dysfunction, growth retardation accelerated thymus huddled, skin atrophy sarcopenia, vascular calcification, osteopenia, elderly lung, cognitive impairment, hearing loss, reduce motor neuron degeneration et al. During the development of chronic kidney disease, chronic kidney disease can lead to the reduction of the klotho gene expression, on the other hand, reduced klotho gene expression will further aggravate the progression of renal disease and the incidence of complications. In the process of acute kidney injury, the expression of klotho gene is unnormal, it is suggested that klotho gene and kidney damage are closely related. With the continuous improvement of people’s living standards, the incidence of diabetes has been increasing, while the incidence of diabetic nephropathy are rapidly improving.Recent animal studies have shown that:dehydrated mice with the change in plasma osmolality have kidney Klotho gene expression changes. The Klotho gene expression was significantly decreased along with the antidiuretic hormone, andaldosterone rised; cells experiments found in relatively constant osmotic pressure, antidiuretic hormone and aldosterone reduced the expression of Klotho gene.it proved that antidiuretichormones as well as the role of aldosterone in the body affect the Klotho gene expression to a certain extent. The body environment is complex, it is osmotic pressure changes affect Klotho gene expression directly or just the antidiuretic hormone and aldosterone levels changes by the changes of osmotic pressure, it still needs further research to confirm. It is considered that diabetic nephropathy mainly affect the renal blood vessels, but whether Klotho gene involved, if it is.how to participate is not very certain. It can be affected by high blood sugar, but whether the hyperglycemia induced osmotic changes that involved are unconcern. This increases of the osmotic pressure affect the kidneys and even multiple organ, but the body is a complex aspect whole cell experiments can not completely simulate the body’s environment, further understanding need to be a large number of clinical trials to prove.Purpose:To explore how high sugar and hypertonicity effect Klotho expression in renal tubular epithelial cells and to study the impact of these two factors is seperate or is a correlation.Methods:Culture rat proximal tubular epithelial cells (NRK-52E), divide the cultured cells into normal control group, high sugar and hypertonic culture group. Extract of Klotho mRN A and protein using RT-PCR and Western blot to detect the Klotho mRNA and protein expression of NRK-52E cells under high-sugar and hypertonicity.Results:(1) RT-PCR results showed that high glucose induced the expression of Klotho mRNA of tubular epithelial cells, hypertonicity also induced the expression of Klotho mRNA of tubular epithelial cells renal, but there were no significant differences both in these.(2) Western blot results showed that high glucose induced the expression of Klotho protein of tubular epithelial cells,hypertonicity also induced the expression of Klotho protein of tubular epithelial cells, but there were no significant differences both in these.Conclusion:Hypertonicity can directly affect the expression of Klotho renal tubular epithelial cells, high sugar can also directly affect the expression of Klotho of renal tubular epithelial cells, but maybe high sugar by affecting the osmotic pressure to affect the expression of Klotho of renal tubular epithelial cell.
