Objective:To study the The Bacteroides thetaiotaomicron intervention intestinalcolonization in diabetic mice and correlation with glucagon-like peptide-1(GLP-1). ToExplore the possible mechanisms of the effects of diabetes. Methods: Cultivation theATCC29148standard strains with BHI blood agar medium,Then plate count andTurbidimetric method Determination of the number of Bacteroides thetaiotaomicron.6-week-old Kunming mice group of blank control group (n=6), high BT group1012Cfu/ml(n=6), medium BT group109Cfu/ml(n=6), and the low BT group106Cfu/ml(n=6),three days of high BT1012Cfu/ml group (n=6), medium dose three days109Cfu/ml(n=6),and three days of low doses of106Cfu/ml(n=6),Determined the number of BT by real timePCR,Determine colonization law of BT.Base on the colonization of Bacteroidesthetaiotaomicron, Intervention in the6-week-old Kunming mice blank control group(n=10), the blank BT (n=10), and intraperitoneal injection of alloxan-made the diabetesmodel Kunming mice model group (n=10) and model BT group (n=10), To observe theweight and fasting glucose level of mice between the15days.Determined the number ofBacteroides thetaiotaomicron by real time PCR; The levels of GLP-1on portal blood andcolon tissue homogenates were measured by enzyme-linked immunosorbent assay (ELISA)to reflect the secretion of GLP-1; To reflect with diabetes in serum C-peptidelevels.Result:1.Bacteroides thetaiotaomicron ATCC29148growth well in BHI bloodagar, have a a single colony, non-polluting. high BT group1012Cfu/ml rating most, astatistically significant difference(P<0.05) compared with the other six groups(P<0.05).2.Groups weight increasing in the15days process, but the blank compared to blank BTgroup, the model is compared to the model BT group, weight increasing is moresignificant (P<0.05). FBG Is normally between blank group and model group, comparedwith the blank, the blank BT group FBG level increased(P<0.05),and The change betweenglucose and BT in stool was positive.3.Model group compared with the blank group, the model BT group compared with the model group, blank BT group compared with blankgroup. the levels of GLP-1on portal blood and colon tissue homogenates were reduced (P<0.05).4.At four groups mice,as BT(Copy number/g)as ordinates, as general indicatorsas abscissa. A: The change between wight and BT was negative(r=-0.70, P<0.05). B:Thechange between FBG and BT was positive(r=0.71, P<0.05).C:The change betweenC-peptide levels and BT was negative(r=-0.62,P<0.05).D:The change between HOMA-IRand BT was positive(r=0.55,P<0.05). E:The change between HOMA-IS levels and BTwas negative(r=-0.43, P<0.05).5.At four groups mice,as BT(Copy number/g) asordinates, as GLP-1leave as abscissa.A:The change between GLP-1on portal bloodlevels and BT was negative(r=-0.73,P<0.05).B:The change between colon tissuehomogenates levels and BT was negative(r=-0.52, P<0.05).Concludes:1.The exogenousadministration of Bacteroides thetaiotaomicron can colonization in the intestine.2.Thesecretion and function of GLP-1in intestinal was related to bacteroides thetaiotaomicron, which prompted that the change in the intestinal flora was concerned with diabetes.
