Changes And The Role Of Th17Cells Before And After The Rat Alveolar Hydatid Excision Surgery

Objective: To determine the expression of Th17cytokines (IL-17, IL-23) betweenbefore and postoperative hydatid lesion resection, and find out the postoperative immunestatus changes. Methods: SD rats were divided into control group, alveolar hydatid group,operation group. They were infected in the groin by injecting with alveolar hydatid (salineinstead of the control group) respectively. The successfully infected rats were included inthe trial after3monthes. Operation group rats were excised the subcutaneous hydatidlesions respectively.6rats were sacrificed after surgery between1to7days. And6rats ofcontrol group and alveolar hydatid were sacrificed on the7th day. The inferior vena cavablood and liver tissue were acquired respectively. Expression levels of IL-17and IL-23inserum and liver homogenates organizations were measured by ELISA, and IL-17mRNAand IL-23mRNA were detected using Real-time RT-PCR. Results: Serum IL-17andIL-23of hydatid group were lower than the normal group. After the surgery, respectivelybegan to increase in the2nd day and3rd day, to tended to be normal in6th day7th day.Liver homogenates IL-17and IL-23of hydatid group were lower than the normal group.After the surgery, respectively began to increase in the3rd day and4th day, IL-17beganto tended to be normal in7th day. IL-23was still significantly lower than the normalgroup in7th day. Liver homogenates IL-17mRNA and IL-23mRNA of hydatid groupwere lower than the normal group. After the surgery, respectively began to increase in the1st day and3rd day, to tended to be normal in4th day6th day. Conclusion: IL-17andIL-23as functional cytokines of Th17cells, as time went on after excision of parasiticAlveolar hydatid, hydatid–induced inhibited state of IL-17and IL-23began to recoverand tends to normal in a relatively short time. which may in turn has become a normalimmune rejection of other infection or an organ transplant.
