objective: to study the epithelial sodium channel alpha subunit gene (alpha ENaC)G2139A, A334T, T663A polymorphic loci and kazak in xinjiang, the Uyghur thecorrelation of primary hypertension. Methods:(1) the case-control of associated researchmethods, to collect1344cases kazak and Uighur primary hypertension group (672cases)and control group (672cases) as the research object, carries on the clinical data ofinvestigation, and collect blood samples.(2) to alpha ENaC) gene sequencing, useTapMan probe PCR-RFLP technique for genotyping.(3) using the Data Analyser andHaploview linkage disequilibrium analysis software, using Phase2.0haplotype analysis.(4)analysis using SPSS18.0statistical software for statistical processing, calculation of twogroups of genotype frequency to confirm compliance with Hardy weinberg equilibrium;Compare two groups of clinical data using t test; Group between genotypes and allele andhaplotype frequencies compared by chi-square test. Results:(1) clinical data analysis ofkazak blood pressure higher than Uyghur, obesity is two national hypertension risk factorsof the disease.(2) application of TaqMan technology and PCR-RFLP method on selectedsamples of alpha ENaC3polymorphic loci gene amplification and genotyping. Results thepolymorphism in kazak, exist in the Uyghur population in xinjiang, and G2139A A334T,T663A genotype frequency distribution conforms to all Hardy Weinberg equilibrium.(3) asingle locus correlation analysis results: alpha ENaC gene G2139A, A334T T663Apolymorphic loci and the distribution of genotype frequency and allele frequency in thetwo ethnic groups in xinjiang in the case group and the control group no difference.(4)linkage disequilibrium analysis results: for the sequencing of the9SNPs were pairedlinkage disequilibrium analysis, the results using Haploview software found in kazak andUyghur groups only G (2139) A and G (+49) C, A(26622) G andT663A there is strongbetween LD, further using Haploview software screening out three tSNPs:(G2139A A334T, T663A).(5) a single body building and site haplotype research results in the casesof the control group: three polymorphic loci, eight kinds of haplotype was established inthe kazak population: AGA, AAG, GGA, GAG haplotype distribution differences in casegroup and the control group, difference has statistical significance (P <0.05); In uyghurpopulation: AGA, AAA, GGG, GGA, GAG, the GAA haplotype distribution is different inthe case group and the control group, the difference had statistical significance (P <0.05);And distribution between the two peoples have difference (P <0.05). Conclusion:(1) thekazak population blood pressure higher than Uyghur, obesity is two national hypertensionrisk factors of the disease, smoking and age is closely connected with two nationalessential hypertension;(2) alpha ENaC gene G2139A, A334T and T663A single locusmay has nothing to do with the xinjiang kazak and uyghur essential hypertension;(3)multiple loci conjoint analysis found that haplotype frequency in the two ethnic groups inxinjiang there are differences in the distribution of cases in the control group, the hint ofthree loci of haplotype associated with two national essential hypertension;(4) haplotypefrequency distribution is different in the two nationalities, suggest that gene polymorphismloci ethnic specificity of distribution.
