Objective: This study is focused on the intervention of resveratrol onapoptosis induced by cisplatin in renal tubular epithelial cells.Methods: Strain HK-2of renal tubular epithelial cell was propagated invitro. The experiments included the normal control group、cisplatin exposure groupand various concentration resveratrol treatment group. The cisplatin-mediatedcytotoxic effects on HK-2were determined by Lactate dehydrogenase releaseexperiments. At the same time, the level of NAG was detected both before and aftercisplatin or cisplatin added with various concentrations of resveratrol treatment.Cell proliferation was monitored through MTT analysis. Morphological changes ofcells were assessed by Hoechst staining. And the apoptosis in HK-2cells wereanalyzed by Annexin V-FITC/PI flow cytometry.Results:1. Lactate dehydrogenase release experiment showed that0~12.5μg/mL ofcisplatin had no obvious influence on HK-2cells (P>0.05). When HK-2cells werecultured with12.5μg/mL cisplatin and different concentrations of resveratrol for24hours,0~0.1mmol/L resveratrol had no obvious influence on HK-2cells(P>0.05).Compared with the control group,0.5mmol/L resveratrol led to releasingof LDH increased markedly(P<0.05).2. MTT analysis suggest that12.5μg/mL cisplatin can inhibit markedly HK-2cells the values of OD (P<0.01). After various concentrations of resveratroltreatment, the values of OD were increased with the concentrations growingup(P<0.05). 3. After the HK-2cells were treated with12.5μg/mL cisplatin, compared withthe control group, the level of NAG increased markedly, the difference wasconsidered statistically significant (P<0.5). And resveratrol significantly decreasedcisplatin-induced release of NAG in HK-2cells (P<0.05).4. The results of Hoechst staining showed that cisplatin could induce the HK-2cells apoptosis, and brought about the formation of apoptosome. However,resveratrol could inhibit the HK-2cells apoptosis in some degree (P<0.05).5. Resultts of flow cytometry demonstrated that cisplatin significantly inducedHK-2cells apoptosis(P<0.01).After the HK-2cells were treated with variousconcentrations of resveratrol,compared with the control,the apoptosis rationotablely declined in a dose-dependent manner.Conclusions:Resveratrol have the capability of suppressing apoptosis of HK-2cells in atime and dose-dependent manner. Thus, Resveratrol may play a protective role forthe renal tubular epithelial cells.
