| ã€Objectives】Accordingly, we used HepG-2cells treated with43℃-cultured as a thermotolerancecells model called (HePG-2/thermotolerance,HePG-2/tt)) to explore whether HepG-2/tt cells produced MDR,to investigate the changes of endogenous hrdrogensulfide,expression and activity of hrdrogen sulfide synthase.ã€Methods】The Cell model in common hepatic carcinoma HepG-2, at43℃water bathenvironment developing heat-resistant hepatoma cell lines established after4hours(HepG-2/tt).HepG-2cells and HepG-2/tt cells into two groups, and then put them inHepG-2cell and HepG-2/tt cell cultivation of water environment of43degreesCelsius in4hours by trypan blue staining (Trypan Blue dye exclusion) detection ofHepG-2and HepG-2/tt cells;Hoechst33258dyeing, PI apoptosis flow cytometrydetection rate;Methylene blue color spectrophotometer method in detection of HepG-2cell and HepG-2/tt cell supernatant and H2S content H2S synthase activity;WesternBlot detection of HepG-2and CBS, CSE and MPST expression in HepG-2/tt cells.ã€Results】HepG-2and HepG-2/tt, HepG-2cell survival is a survival rate to72.6%±26.3%±0.8%,HepG-2/tt, difference statistically compared the two groups (P<0.01).HepG-2and HepG-2/tt, HepG-2cells apoptosis of42.3%±27.2%±2.6%,HepG-2/tt apoptosis rate. Difference statistically compared the two groups(P<0.01).HepG-2cell and HepG-2/tt cell with two sets of cell supernatant H2S content of hydrogen sulfide content in the supernatant from HepG-2are0.08±(nmol/mg.Protein), HepG-2/tt hydrogen sulfide content in the supernatant were0.143±(nmol/mg.protein)。Difference statistically compared the two groups (P<0.01).HepG-2cell andHepG-2/tt cell with two sets of cells in the H2S synthase, H2S synthase activity inHepG-2cells of0.309±(nmol/min/mg.Protein), H2S synthase activity in HepG-2/ttcells was0.549±(nmol/min/mg.protein)。Difference statistically compared the twogroups (P<0.01).HepG-2cells and expression of HepG-2/tt cells in the CBS, CSE and MPST,heat-resistant strains of CBS, CSE, MPST expression significantly higher (P<0.01).These results suggest that endogenous hydrogen sulfide may be involved in heat-resistant thermal tolerance of the liver cancer cell HepG2/tt.ã€Conclusions】Endogenous H2S may be involved in the thermaltolerance of HepG-2. |
