Objective: In this study, we used9kinds of biomarkers of tumor combined withPSA in test of normal and prostate cancer person to find out that we can set up adiagnostic model for Chinese population or not. We also observed the relationshipbetween these biomarkers and Gleason score to see whether they can aid in theprognosis and tumor grade malignancy.Materials and methods: In our study, there are total388blood serums whichinclude140prostate cancer patients,96benign prostatic hyperplasia patients, and152healthy controls. We first detect the expression of these biomarkers in the blood serumby ELISA method and Luminex method. Then we validate the positive samples byWestern blot. We use the statistic software of SPSS19.0to analyze the data. First wecompare the prostate cancer related diagnostic value of each marker by logisticregression analysis and ROC curve, and then select powerful ones and combined themto screen prostate cancer. We also analyze its sensitivity and specificity to set up amulti-marker diagnostic model. In the end, we analyze the relationship between thesebiomarkers and Gleason score by logistic regression to see whether they can aid in theprognosis and tumor grade malignancy.Result:1. The positive rate of XAGE-1b、LEDGF、NY-ESO-1、SSX-2、CEA、AMACR、CIP2A、HMGB1、AKAP4expressed in PCa patients are:53.57%、47.86%、33.81%、34.53%、23.91%、28.06%、19.12%、32.14%、44.78%,We found thatXAGE-1b、SSX-2、AMACR and AKAP4were highly expressed in Chinese prostatecancer patients.2.The AUC of the four biomarkers plus PSA were0.666、0.615、0.551、0.768、0.672 respectively;The AUC of the combination of the4biomarkers was0.838;The AUCof the four biomarkers combined with PSA was0.888.The sensibility and specificityof the combination of the four biomarkers in diagnose prostate cancer were80%and82.24%respectively.3.(1)In those patients whose PSA value was between4-10ng/ml, the ACU ofXAGE-1b、SSX-2、AMACR、AKAP4and PSA were0.64、0.512、0.533、0.770、0.505;The AUC of the combination of the four biomarkers was0763;The AUC of thefour biomarkers combined with PSA was0763.(2)In those patients, the sensibilityand specificity of the combination of the four biomarkers in diagnose prostate cancerwere83.67%and73.17respectively.4. We found that all the nine biomarkers were not correlated with PSA value andAMACR and NY-ESO-1was significantly correlated with Gleason Score(P<0.05).Conclusion:1.XAGE-1b、SSX-2、AMACR、AKAP4were highly expressed inChinese prostate cancer patients.2.The sensibility and specificity of the combination of the four biomarkers indiagnose prostate cancer were80%and82.24%respectively, which is significantlyhigher than that of PSA. In those patients whose PSA value was between4-10ng/ml,the sensibility and specificity of the combination of the four biomarkers in diagnoseprostate cancer were83.67%and73.17respectively, which is also significantly higherthan that of PSA.3.AMACR and NY-ESO-1was significantly correlated with Gleason Score.
