Objective:To detecte the proportion of CD4~+T cell subsets in peripheral blood of allergicrhinitis(AR) patients and healthy controls, and to explore their significance in thepathogenesis of AR.Methods:Peripheral blood mononuclear cells(PBMCs) of19AR patients and19healthycontrols were isolated. The proportion of CD4~+CD25~+T cells, CD4~+CD25~+Foxp3~+Tcells, IL-4~+CD4~+T cells, IFN-γ~+CD4~+T cells, IL-10~+CD4~+T cells and TGF-β~+CD4~+Tcells in CD4~+T cells of peripheral blood were detected by flow cytometry. Andlymphocyte transformation test was conducted. And the correlation betweenlymphocyte transformation rate and the proportion of CD4~+CD25~+T cells,CD4~+CD25~+Foxp3~+T cells, IL-4~+CD4~+T cells, IFN-γ~+CD4~+T cells, IL-10~+CD4~+T cells,TGF-β~+CD4~+T cells in CD4~+T cells of peripheral blood were analyzed.Results:1. Compared with healthy controls, the proportion of CD4~+CD25~+T cells inperipheral blood CD4~+T cells of AR patients was significantly decreased [(4.11±0.86)%vs.(6.87±0.93)%,t=﹣9.48,P<0.01]; the proportion of CD4~+CD25~+T cells inperipheral blood PBMCs of AR patients was significantly decreased [(0.84±0.38)%vs.(1.48±0.53)%,t=﹣4.27,P<0.01]. Compared with healthy controls, the proportionof CD4~+CD25~+Foxp3~+T cells in peripheral blood CD4~+T cells of AR patients wassignificantly decreased [(2.63±1.22)%vs.(5.24±1.60)%,t=﹣5.66,P<0.01)]; the proportion of CD4~+CD25~+Foxp3~+T cells in peripheral blood PBMCs of AR patientswas significantly decreased [(0.56±0.39)%vs.(1.16±0.62)%,t=﹣3.57,P<0.01].2. Compared with healthy controls, the proportion of IL-4~+CD4~+T cells in peripheralblood CD4~+T cells of AR patients was increased [(11.00±3.88)%vs.(3.98±1.47)%,t=7.38, P<0.01)];3. Compared with healthy controls, although the proportion of IFN-γ~+CD4~+T inperipheral blood CD4~+T cells of AR patients was decreased, there was no statisticallysignificant [(13.59±6.42)%vs.(16.96±7.78)%, t=﹣1.46,P=0.154)].4. Compared with healthy controls, the proportion of IL-10~+CD4~+T cells inperipheral blood CD4~+T cells of AR patients was significantly decreased [(1.66±0.48)%vs.(3.82±0.92)%,t=﹣9.08,P<0.01)].5. Compared with healthy controls,the proportion of TGF-β~+CD4~+T cells inperipheral blood CD4~+T cells of AR patients was significantly decreased [(1.92±0.54)%vs.(4.76±1.12)%,t=﹣9.94,P<0.01)].6. Compared with healthy controls, the lymphocyte transformation rate in peripheralblood of AR patients was significantly increased [(62.32±12.71)%vs.(49.84±6.53)%,t=3.804,P=0.001)].Lymphocyte transformation rate was negatively correlated with theproportion of CD4~+CD25~+Treg cells in CD4~+T cells(r=﹣0.507, P=0.001); Lymphocytetransformation rate was negatively correlated with the proportion of CD4~+CD25~+T cellsin PBMCs(r=﹣0.464, P=0.003); Lymphocyte transformation rate was negativelycorrelated with the proportion of CD4~+CD25~+Foxp3~+T cells in CD4~+T cells(r=﹣0.397,P=0.014), Lymphocyte transformation rate was negatively correlated with theproportion of CD4~+CD25~+Foxp3~+T cells in PBMCs(r=﹣0.368,P=0.023);Lymphocytetransformation rate was positively correlated with the proportion of IL-4~+CD4~+T cells inperipheral CD4~+T cells(r=0.350,P=0.031); There was no significant correlationbetween lymphocyte transformation rate and the proportion of IFN-γ~+CD4~+T cells inperipheral CD4~+T cells (r=﹣0.298,P=0.069); Lymphocyte transformation rate was negatively correlated with the ratio of IL-10~+CD4~+T cells in peripheral CD4~+T cells(r=﹣0.420,P=0.009); Lymphocyte transformation rate was negatively correlated with theproportion of TGF-β~+CD4~+T cells in peripheral CD4~+T cells(r=﹣0.454,P=0.004).Conclusions:1. The proportion of CD4~+CD25~+T cells and CD4~+CD25~+Foxp3~+T cells in peripheralblood was significantly decreased, suggesting that the reduction in the proportion ofCD4~+CD25~+Treg cells may be an important factor in the pathogenesis of AR.2. The proportion of IL-4~+CD4~+T(Th2) cells was significantly, and IL-4can resist theimmunosuppressive function of Treg cells, which resulting in dominant Th2-typeimmune response. Therefore, IL-4may be an important therapeutic target of AR.3. The proportion of IFN-γ~+CD4~+T(Th1) cells decreased, not statistically significant.May be due to the characteristics of IFN-γ,which has both pro-inflammatory andsuppression of inflammatory response.4. A lower proportion of IL-10~+CD4~+(Tr1) cells may be an important factor in thepathogenesis of AR. Improve the proportion of IL-10secreting Tr1cells may be of greatsignificance in the treatment of AR.5. The proportion of TGF-β1~+CD4~+T(Th3) cells in peripheral blood was significantlydecreased, which may be may be an important factor in the pathogenesis of AR.However, few studies about the relationship between TGF-β1and AR, especially therelationship between TGF-β1levels in peripheral blood and AR, further research isneeded.6. The study also indirectly suggest that CD4~+CD25~+Treg cells, IL-10~+CD4~+T(Tr1)cells, TGF-β1~+CD4~+T(Th3) cells with immune suppression ability. WhileIL-4~+CD4~+T(Th2) cells with the promotion of cell-mediated immunity ability.
