This thesis is composed of four chapters. In the first chapter, the chemical constitunents from70%methanol extract of the leaves aromatic tobacco were studied. The second chapter described Fourier Tansform Near Infrared Spectroscopy stimultaneous determination of chlorogenic acid, rutin, scopoletin and total polyphenol in tobacco. The third chapter described RP-HPLC stimultaneous determination of solanesol in Tobacco Leaves. The last chapter summarized Nicotiana’s chemical constituents and pharmacological actions.By various kinds of chromatographic separation methods,30compounds were isolated. On the basis of physical-chemical properties and spectral data, they were identified, including eighteen flavones, twelve phenylpropanoids. They are respectively:Nicotpanoid A (1*), Nicotpanoid B (2*),6,7-dimethoxy-4’-hydroxy-8-Formylflavon (3*),8-formyl-4’,6,7-trimethoxyflavon (4*),4’,7-dihydroxy-8-formyl-6-methoxyflavon (5*), Kaempferol (6), Kaempferol-3-O-β-D-gl-ycosides (7), Kaempfe-rol-3-O-β-D-galactopranoside (8), Quercetin (9), Quercet-in-3-O-β-D-glycosides (10), Quercetin-3-O-β-D-galactopranoside (11), Rutin (12), Diosmetin-7-rutinoside (13), Taxifolin (14), Taxifolin4’-methyl ether (15),(-)-epicatech-in (16), Morachalc-one A (17), Chlorogenic acid (18), Chlorogenic acid methylester (19),4-O-caffeoylq-uinic acid (20),(±)-Pinocembrin(21),5,7,4’-Trihydroxyflavanone(22),5-O-caffeoylq-uinic acid (23), Kaempferol-3-O-β-D-rutinsides (24), Caffeic acid (25), Ferulic acid (26),(E)-ferulaldehyde (27), coumaric acid (28), ω-Hydroxypropioguaiacone (29),3-methoxy-4-hydroxy-propyl alcohol (30). Among them, compounds1-5are news, and they were carried out bioactive screening test. As a result, they showed significant inhibitory effect against human tumor cell lines.To establish the NIR spectroscopy prediction model of chlorogenic acid, rutin, scopoletin and total polyphenol by Fourier Tansform Near Infrared Spectroscopy. In the range of7500to4000cm-1,partial least square (PLS) regression method, second derivative and Norris derivative filter were used.The content of solanesol in tobacco materials and tobacco products was determined by reverse phase high performance liquid chromatography. A waters C18column (4.6mm×250mm,5μm) was used with a mobile phase of methanol-ethanol (75:25) mixed solvent at the flow rate was1.0mL-min-1,and the detective wavelength was set at211nm.Not only did the natural chemical constituents isolated, but also the pharmacological investigation of active components in tobacco in this thesis.
