Objective Estabish mechanical disturbance of spermatogenesis models of rabbits and observe spermatogenic effect of Guiqishengjing capsule on these rabbits models by analyzing the changes of rabbits semen parameters, testicular pathology which provides experimental basis for traditional chinese medicine for the treatment of male infertility.Method Use the surgical method to establish rabbit model of unilateral (male Japanese white rabbits) and bilateral (male California rabbits)testicular spermatogenic impairment, and different subgroups was divided according to whether perform testicular descent fixation or not. Huangjingzanyu capsules and warm water was considered as control group, then we observe the influence of Guiqishengjing capsule to rabbit sperm density, viability, motility and semen volume through vertical and horizontal comparison.Results1. Unilateral dyszoospermia model in male Japanese white rabbits:1.1There were significant statistical difference in sperm density, motility, and viability on the groups whether perform testicular descent fixation or not before and after unilateral modeling through vertical comparison (all P<0.05). But no significant statistical difference was found in semen volume (all P>0.05) and there were significant pathological changes in testicles of different groups.1.2There were significant statistical difference in sperm density, motility, and viability (all P<0.05) and pathological changes in testicles but no difference in semen volume (all P>0.05) compared the subgroups of performing testicular descent fixation (a1,a2,a3,c1,c2,c3) with the subgroups of no testicular descent fixation (b1,b2, b3,d1,d2,d3) at fourth month postoperatively.1.3In the fourth month after unilateral testicular descent fixation, pairwise comparisons of sperm density, motility, and viability were performed among the subgroups a1, a2, a3, and among c1, c2, c3.There was no significant statistical difference between a2and a1, c2and c1, a3and a1,c3and c1(all P<0.05),while no significant statistical difference was found between a3and a2, c3and c2.(all P>0.05). And there was no significant difference in semen volume among these different subgroups (all P>0.05).In subgroups with no testicular descent fixation, pairwise comparisons of sperm density, motility, and viability were also performed, but no significant statistical difference was found (all P>0.05).2. Similarly, There were significant statistical difference in sperm density, motility, and viability(all P<0.05) but no difference in semen volume (all P>0.05) between subgroups with unilateral testicular descent fixation (B1,B2,B3, C1,C2,C3) and subgroups with bilateral testicular descent fixation (D1,D2,D3)2.3In the fourth month after bilateral testicular descent fixation, pairwise comparisons of sperm density, motility, and viability were performed among the subgroups B1,B2,B3, and among C1,C2,C3, and among D1,D2,D3. The results shew that significant statistical difference was found between B2and B1,C2and C1,D2and D1,B3and B1,C3and C1,D3and D1(all P<0.05), while there was no significant difference between B3and B2, C3and C2%D3and D2. And we found no significant difference in semen volume among these different subgroups (all P>0.05). In subgroups with no testicular descent fixation, pairwise comparisons of sperm density, motility, viability and semen volume were also performed,but no significant statistical difference was found (all P>0.05).Conclusion1. Temperature factors on rabbit spermatogenic disorders have a certain degree of reversibility.2. Incentive (temperature) caused impaired spermatogenesis is not removed, rabbit semen quality (sperm density, vitality, motility) decreased progressively, testicular histopathological changes continue to increase.3. Incentive (temperature) caused impaired spermatogenesis were removed and then give the seminiferous medication can better promote the recovery of the quality of rabbit semen (sperm density, viability, motility) and testicular histopathological changes, Guiqishengjing capusle and Huangjingzanyu capsule which has been listed have a good curative effect It lays the foundation for the future research for mechanism and clinical trials of Guiqishengjing capsule.
