Objective:Detect the content of the growth factor in serum in the rats femur fracture’ model in different time, explore the relationship between expression changes of growth factor and different injury time establish a new method of fracture timing surmise, and provide experimental basis for the judgement of clinical healing of fracture.Methods:36healthy male SD rats were randomly divided into control group, sham group and experimental group (n=12). For the experimental group, rats are received open osteotomy on the right midshaft of right femur.Then the osteotomies were immobilized by retrograde Kirschner wire. For the sham group:the skin and muscle of right legs of the rats were cut and then sutured. Each animal took orbital venous blood samples1.5ml at1st,3rd,5th days and1st,2nd,3rd,4th,8th,12th weeks after the operation. Detect the serum level of TGF-β1, bFGF, PDGF, EGF, NGF with the method of double antibody sandwich ELISA, measure the absorbance values and calculate the content of corresponding growth factor of all samples, statistical analysis.Results:1. The serum concentration of TGF-β1significantly increased in the third day after the fracture surgery.Tt doubled in the second week and began to dorp in the third week and forth week, but was still higher than the level of the blank groups’. Compared with the control group and the sham surgical group, the serum concentration of TGF-β1in fracture group has significant difference (P<0.05).The serum concentration of TGF-β1change of three groups were statistically insignificant after8weeks (P>0.05).2. The serum concentration of bFGF began to rise in the third day after the fracture and reached a peak in the second week. The serum concentration of bFGF dropped in the third week and the forth week. Compared with the control group and the sham surgical group, the serum concentration of bFGF in fracture group has significant difference (P<0.05). The serum concentration of bFGF change of three groups were statistically insignificant after8weeks (P>0.05).3. The serum concentration of PDGF began to rise in the first day after the fracture and reached a peak in the second week, and began to drop in the third week. Compared with the control group and the sham surgical group, the serum concentration of PDGF in the fracture group has significant difference (P<0.05). The serum concentration of PDGF change of three groups were statistically insignificant after4weeks (P>0.05).4. The serum concentration of EGF began to rise in the first day after the fracture and reached its peak in the fifth day, and began to drop in the first week. It was higher than the normal level in the second week and third week. Compared with the control group and the sham surgical group, the serum concentration of EGF of the fracture group has significant difference (P<0.05). The serum concentration of EGF change of three groups were statistically insignificant after4weeks (P>0.05).5. The serum concentration of NGF began to rise in the first day after the fracture, and reached a peak in the second week, and began to drop in the third week. Compared with the control group and the sham surgical group, the serum concentration of NGF of the fracture group has significant difference (P<0.05).The serum concentration of NGF change of three groups were statistically insignificant after4weeks (P>0.05).Conclusions:1. The serum concentration of TGF-β1and bFGF rose after the fracture surgery in the third day and reached a peak in the second week. It then gradually began to drop and was reduced to normal in the8th week. The serum levels of TGF-β1and bFGF can be used to surmise time of fracture, has greater value for2weeks of fracture surmise.2. The serum concentration of PDGF and NGF began to rise after the fracture surgery in the first day and reached a peak in the second week. Then gradually dropped and reduced to normal in the forth week. The serum levels of PDGF and NGF can be used to surmise fracture time, has greater value for2weeks of fracture surmise.3. The serum concentration of EGF began to rise after the fracture surgery in the first day and reached a peak in the second week. It then gradually dropped and was reduced to normal in the forth week. The serum levels of EGF can be used to surmise fracture time, has greater value for1weeks of fractures surmise.4. TGF-β1、bFGF、PDGF、EGF、NGF that Five Cytokines joint detection has important value for2weeks of fracture surmise.
