ObjectiveThis study is to investigate expression of ras-related C3botulinum toxin substrate1(Racl)and NF-κB p65in Non-Small Cell Lung Cancer(NSCLC) and their clinical significance.MethodsImmunohisto-chemical staning was applied to detect expression of Racl and NF-κB p65in NSCLC group (112cases) and control group(112cases) who underwent operations during May2007to June2008. Differences of expression of Rac1and NF-κB p65in NSCLC tissues and normal lung tissues were analysed; correlations of expression of Racl and NF-κB p65to clinical pathological parameters and prognosis and their correlations were analysed.Results(1) The expression intensities of Racl(88.39%VS19.64%) and NF-κB p65(90.18%VS8.04%) in NSCLC were distinctly higher than those in control group.The expression of Rac1and NF-κB p65in NSCLC demonstrated significantly positive relation(r=0.656P<0.001).(2) The expression of Racl and NF-κB p65in NSCLC was not significantly different in Gender, age, smoking status, tumor location, tumor size and histological type(P>0.05).(3)The expression of Racl in NSCLC was significantly different in differentiation, TNM stage and lymph node metastasis(P<0.05or P<0.01).The expression of NF-κB p65in NSCLC was significantly different in TNM stage and lymph node metastasis(P<0.05or P<0.01). What’s more, the "coordinated expression" of Racl and NF-κB p65in NSCLC was significantly different in differentiation, TNM stage and lymph node metastasis(P<0.05or P<0.01).(4)The3-year survival rates were lower in patients with Racl high expression(35.29%) than those with low expression(79.07%) while the3-year survival rates were lower in patients with NF-κB p65high expression(40.28%) than those with low expression (74.36%).furthermore, the3-year survival rates were lower in patients with both Racland NF-κB p65high expression(37.70%) than those with either Rac1or NF-κB p65high expression(61.11%) and those with both Rac1and NF-κB p65low expression (81.25%).(5)Further multivariate analysis suggested that both high expression of Racl and NF-κB p65and lymph node metastasis were prognostic indicators for NSCLC(P<0.05,P<0.001).ConclusionThe expression of Racl in correlates with the expression of NF-κB p65in NSCLC, which contributes the malignancy-related of NSCLC. Detecting the expression of Racl combined with NF-κB p65in NSCLC tissue may give a clue on prongnosis of patient.
