Objective:Degeneration and angiogenesis of intervertebral disc are basic pathological changes of IDD (Intervertebral disc degeneration).TGF-β1play an important role in the reduce of matrix proteoglycan content. There are few research about the relationship between TGF-β1and angiogenesis. We want to to investigate the expression of TGF-β1in the intervertebral disc degeneration and its relationship with angiogenesis by immunohistochemistry.Materials and methods:We collected38cases formalin-fixed, paraffin-embedded tissues of IDD patients during January2009to October2011in Tianjin hospital. We also selected10cases of normal disc tissue as the control group. The tissues ware section thicked4μm. We examinated the expression of the antibodies of TGF-β1,CD34in IDD and normal disc tissues with immunohistochemistry En Vision method.(The datas were analyzed by x±s and compared with independent-samples T test and paired-samples T test by SPSS17.0).Statistical significance was defined as P<0.05.Results:1. The expression of TGF-β1in degenerative intervertebral disc tissue and in normal intervertebral disc tissue:The positive expression of TGF-β1in38cases of degenerative intervertebral disc tissue positive is25cases, the positive rate is65.8%(25/38). Within normal intervertebral disc tissue we have not seen TGF-β1positive expression. The former is significantly higher than the latter, the difference was statistically significant (P<0.05).2. The positive rate of TGF-β1in three-pathological-type IDD was obviously difference(P<0.05):prominent group of disc tissue15.4%(2/13), prolapse group of disc tissue90.0%(9/10), free group disc tissue93.3%(14/15). the difference was statistically significant (P<0.05).3. The relationship between positive rate of TGF-β1and pain scores:grade I73.0%(19/26), grade Ⅱ50.0%(6/12). the difference was statistically significant (P<0.05). 4. There was no relationship between the expression of TGF-β1and clinical pathological parameters (P>0.05),but the duration of low back pain(P<0.05).5. The expression of CD34in degenerative intervertebral disc tissue and in normal intervertebral disc tissue:The positive expression of CD34in38cases of degenerative intervertebral disc tissue positive is24cases, the positive rate is63.2%(24/38). Within normal intervertebral disc tissue we have not seen CD34positive expression. The former is significantly higher than the latter, the difference was statistically significant (P<0.05).6. The positive rate of CD34in three-pathological-type IDD was obviously difference(P<0.05):prominent group of disc tissue30.8%(4/13), prolapse group of disc tissue80.0%(8/10), free group disc tissue80.0%(12/15). the difference was statistically significant (P<0.05).7. The relationship between positive rate of CD34and pain scores:gradeⅠ84.6%(22/26), grade Ⅱ50.0%(6/12). the difference was statistically significant (P<0.05).8. There was no relationship between the expression of CD34and clinical pathological parameters (P>0.05),but the duration of low back pain(P<0.05).9. There was significantly positive correlation between TGF-β1and CD34(P<0.05).Conclusion:1. The TGF-β1positive expression rate in IDD patients was obviously higher than normal group, TGF-β1may play an important role in the IDD pathogenesis,2. The TGF-β1positive expression rate in prolapse group of disc tissue and free group disc tissue was obviously higher than prominent group of disc tissue, TGF-β1may play an important role in the absorption of IDD.3. The study on the relationship of TGF-β1and CD34may be can reveal the mechanisim related to angiogenesis and provide theoretical evidence for therapy.
