Purpose:To culture humanized prostate cancer cancer cell line DU145in vitro and establish subcutaneous xenografts models of living nude mice;By observing18F-FDG PET/CT imaging after argon-helium cryosurgical ablation of the prostate cancer xenografts in nude mice and comparing these features and quantitative analysis variations with tumor size,cell morphology,tumor cell proliferation,cell apoptosis and so on,evaluate early the feasibility and role of18F-FDG PET/CT imaging in xenografts in argon-helium cryosurgical ablation in vivo conditions and the molecular level and thereby supply scientific theory for further clinical applications.Methods:(1)Culture humanized prostate cancer cancer cell line DU145in vitro.When the cells grew to exponential growth phase, establish the prostate cancer subcutaneous xenograft nude model by the injection of tumor cell suspension and small xenograft.Observe the growth situation of prostate cancer subcutaneous xenograf and count the volume of them and draw their growth curve;(2)Via tail intravenous injection about100μci dose every prostate cancer subcutaneous xenograft nude,18F-FDG PET/CT imaging was applying after1h or so and observed;(3)18mice were randomly divided two group(the treatment group and the control group).Every group had9mice.The treatment group had argon-helium cryosurgical ablation of the prostate cancer xenografts in nude mice.The control group didn’t have any therapy of the prostate cancer xenografts in nude mice.The treatment group were conducted18F-FDG PET/CT imaging after argon-helium cryosurgical ablation of the prostate cancer xenografts. The control group were conducted18F-FDG PET/CT imaging before argon-helium cryosurgical ablation of the prostate cancer xenografts in nude mice of the treatment group. Observe their tumor size and cell morphology. Their metabolic activity T/NT values(tumor lesions and normal tissues of the SUVmax value ratio, that is,T/NT values),tumor cell proliferation and cell apoptosis were analyzed by Excel and SPSS statistical software packages17.0. Evaluate early the role of18F-FDG PET/CT imaging in prostate cancer xenografts after argon-helium cryosurgical ablation in vivo conditions and the molecular level.Results:(1)Establish successfully humanized prostate cancer DU145cell subcutaneo- -us xenografts models of living nude mice and draw their growth curve.The subcutaneous xenografts began to grow rapidly about20day later. Their diameter of the subcutaneous xenografts was lcm after more than40days (2)Via tail intravenous injection,18F-FDG PET/CT Imaging was applying after lh or so and observed.18F-FDG uptake was higher in prostate tumors and a kind of positron imaging agents which evaluates the role of argon-helium cryosurgical ablation;(3)After argon-helium cryosurgical ablation, the xenografts of the treatment group had lower18F-FDG uptake;(4) After argon-helium cryosurgical ablation, immunohistochemical index of the xenografts of the treatment group began to change.Tumor cell proliferation were restrained and cell apoptosis were induced.Conclusions:(1)Establish successfully humanized prostate cancer DU145cell subcutaneous xenografts models of living nude mice;(2)18F-FDG PET/CT can detect and measure their metabolic activity in xenografts after argon-helium cryosurgical ablation in vivo conditions and the molecular level,evaluate effectively and early the role of argon-helium cryosurgical ablation and supply scientific theory for further clinical applications.
