Platycodon gradiflorum (Jacq.) A.DC. can treat asthma and bronchitis when as a traditional Chinese medicine, and it is also as favorite food in many countries. In the recently reports, separation and pharmacological activities of platycodin were paid much attention. But there were few researches on the sapogenin.In this study, platycodin was extracted by shanshi extractor. After passed through AB-8resins, platycodin was concentrated and then hydrolysis with acid. In order to screening the optimum process, a series of single factor experiments were carried out. The single factor included that volume fraction of sulfuric acid, the temperature, the period and times of hydrolysis. Based on the results of the above experiments, an orthogonal test was designed. The results showed that the stable and reliable process as follows:volume fraction of sulfuric acid was8%, temperature was90℃, the period was8h and3times.The hydrolyzate was dissolved in ethyl acetate. One compound was gained by the method of thin-layer chromatography, column chromatography on silica gel and HPLC repeatedly. It was identified as polygala acid by’H-NMR,13C-NMR and IR.A model was made by the method of inoculated HepA22hepatoma cells in the left forelimb armpit of mice. The antitumor effect of sapogenin was conducted in vivo. The mice that inoculated successfully were divided into four groups, as follows:model group, positive group, low and high dosage of sapogenin(N=10in each group). Control and model group were received5%o CMC-Na. Positive group was received cyclophosphamide (20mg/kg).30mg/kg and50mg/kg were the low and high dosage respectively. After12-day treatment, there were decreases in the groups of sapogenin, while inhibition rate were50%and56%, respectively.The antioxidant effects of sapogenin were determined by the methods of O2-radical and DPPH radical. The O2-scavenging ability of sapogenin was the best when0.2mg/mL, the scavenging rate was75.96%. While the DPPH scavenging ability of sapogenin was the best when0.4mg/mL, the scavenging rate was63.35%. In antibacterial tests, sapogenin had a good inhibition against Staphylococcus aureus and Escherichia coli when40mg/ml, and had weaker inhibition on Salmonella typhimurium and Pesudomonas pyocyaneum.
