Objective:To investigate the effects of Quercetin on the expression of serum resistin and insulin resistance(IR) in rats with non-alcoholic fatty liver disease(NAFLD). Methods:The SD rats were treated with different dose of quercetin by gavage for next8weeeks, after they were raised with high-fat diet for4weeks. The serum Resistin, serum triglyceride (TG), fasting plasma glucose (FPG), fasting insulin (FINS) and malondialdehyde (MDA) were determinded before and after the administration of quercetin. And insulin resistance index (HOMA-IR) was calculated. Moreover, the pathological changes of the liver were observed. Results:The results of pathological examination showed that the degree of degeneration was decreased in the high and low dosages of quercetin comparing with the mode group,which u value of4.41'2.19,P<0.05, the difference was statistically significant.And pathological degree was lighter in the high-dose group than that in the low-dsoe one (u=2.44, P<0.05). The expressions of the high-dose group serum resistin (ng/ml)、FPG(mmol/L)、FINS(mU/L) and HOMA-IR were1.00±0.13、5.34±1.00、4.66±1.84、1.31±0.22, the expression levels of the same indicators of the low-dose group were1.17±0.15,6.90±1.47,8.49±1.96,1.70± 0.26,which were lower than that in the model group which express1.31±0.22、18.92+1.29、10.06±3.44、16.5±5.23、2.01±0.98.Compared with the model group.High and low dose group t value of5.98、10.93、7.45、12.69、4.22'2.70、8.21、4.99、8.591.87respectively,P<0.05,the difference was statistically significant. The amount of serum TG (mmol/L)and liver MDA (nmmol/mgprot) expression levels were0.35±0.06and17.64+5.77,were lower than the model group which express0.48±0.14and24.65+4.85,compared with the model group,t value of4.70and5.14, P<0.05, statistically significant.The amount of serum resistin、FPG、FINS and HOMA-IR decreased in high-dose group compairing with low-dose group,t value of3.28,2.72,2.46,4.10,2.35, P<0.05, statistically significant. Normal group, model group,the high-dose and the low-dose group’s serum resistin, TG, liver MDA correlation analysis with HOMA-IR showed:the resistin groups correlation coefficient r values were0.80、0.89and0.71, TG group correlation coefficient r-values0.93、0.90、0.91and0.86, the correlation coefficient of the liver MDA group r values were0.96,0.67,0.69and0.67. r>0, The correlation analysis showed that the expression of resistin, TG, FPG and FINS were positively correlated with HOMA-IR and the degree of liver disease.Conclusions:Quercetin by reducing the level of serum resistin,TG, FPS, FINS, liver tissure MDA and HOMA-IR, to improve the insulin resistance, inhibition of lipid peroxidation,as well as have effects on levels of inflammatory cytokine in the rats with NAFLD, but also the effect of high dosage of quercetin was more obvious.
