[Objective] To study the reproductive toxicology of fluoride and sodium sulfite alone or combination effect on MAPK signal pathway in testicular tissue in mice, and discuss the cellular and molecular mechanisms of toxicity of fluoride and sodium sulfite to the reproductive organs of male animals, in order to supply scientific evidence for control fluorosis and sodium sulfite to male reproductive toxicology.[Methods] Simulation the environment pollution, healthy male Kunming mice obtained from the Experimental Animal Center of Shanxi Medical University were exposed to sodium fluoride and sodium sulfite following four groups of18mice:Control group; Sodium fluoride group; Sodium sulfite group; sodium fluoride and sodium sulfite combination group. The MAPK pathway (ERK1/2/5, JNK1/2/3and p38) and transcriptional factors of JNK downstream (C-Jun, JunD, Elk-1and ATF-2) expression levels in mice testis were analyzed by Real-time RT-PCR.[Results]1. Compared with the control group, after poison exposure at7week, the expression levels of ERK2mRNA from fluoride group、sodium sulfite group and combination group were decreased29.4%,52.9%(P<0.05) and33.4%(P<0.05)respectively; The expression levels of JNK2mRNA from3treated groups decreased29.6%,59.3%(P<0.01) and56.8%(P<0.01) respectively. However, compared with the control group, ERK1、JNK1、JNK3and ERK5mRNA expression level had no significance in any group.2. Compared with the control group, after poison attack at14week, the expression levels of JNK1from3treated groups decreased39.1%,61.8%(P<0.05) and25.5%respectively. The expression levels of JNK2from3treated groups decreased48.9%,51.2%(P<0.01). The expression levels of ERK5from3treated groups increased85.2(P<0.05),63%, and83%(P<0.05) respectively. However, compared with the control group, ERK1, ERK2JNK3and p38mRNA expression level had no significance in any group.3. Compared with the control group, that the expression levels of C-Jun, MAX, Elk-1and ATF-2mRNA had no significance in any group after poison attack at7week. However, after poison attack at14week, compared with the control group, the expression levels of C-Jun from3treated groups increased44.7%,31.2%, and30.7%(P<0.05) respectively. The expression levels of JunD from3treated groups increased92.1%(P<0.05),36.6%, and150.5%(P<0.001) respectively. [Conclusion] In this research, The ERK1/2/5, JNK1/2/3and p38and transcriptional factors of JNK downstream (C-Jun, JunD, Elk-1and ATF-2) expression levels in mice testis from fluoride and sodium sulfite treated alone or combination were analyzed by Real-time RT-PCR. The results suggested that testis toxicity of fluoride and sodium sulfite was mainly displayed in JNK signaling pathway, and it did affect the transcriptional factors of JNK downstream expression in this study.
