Objective: Recently, our team has discovered the NCAPH gene from genechip technology, which is a markedly abnormal gene in hepatocellularcarcinoma (HCC). We speculate that NCAPH may play an important role in theoccurrence and development of HCC. This study clarifies the influence ofNCAPH gene abnormally high expression on the occurrence and developmentof HCC from the molecular level, in order to provide new ideas for the reveal ofHCC pathogenesis. It even can provide new diagnosis standard for HCC andnew molecular targets for indicators of HCC intervention therapy. Meanwhile,we combined with traditional clinical pathological data, and NCAPH expressionlevel to explore the impact of prognostic correlation factors in hepatocellularcarcinoma patients with surgical removal of the disease-free survival.Methods: Using RT-PCR to detect NCAPH expression levels of HCCtissue and in para-carcinoma tissue of206HCC patients; usingimmunocytochemistry methods to detect NCAPH protein expression levels andthe cellular localization in HCC tissue and in para-carcinoma tissue of206casesof HCC patients; the follow-up clinical data of these patients was collected andorganized. Kaplain-Meier method was used to evaluate survival rates.Log-rank test was used to univariate analysis.Cox’s regression of multivariate analysiswas done in order to determine clinicopathologic factors which had influencehepatectomy HCC patients after hepatectomy.Results: The NCAPH gene positive expression rate was77.18%in thetumor tissue(159/206),the NCAPH protein was highly expressed in71.78%(69/96) of patients with HCC cancer tissue,10.4%(10/96) of patients withpara-neoplastic organization NCAPH protein was highly expressed. Univariateanalysis results show that factors affected the disease-free survival rates of HCCpatients include AFP level, tumor size, number of drinking history, TNM stage,portal vein tumor thrombus formation, tumor metastasis, aspartateaminotransferase level, and NCAPH expression, but the patient’s age, sex,smoking history, family history, cirrhosis, hepatitis B infection, tumor recurrenceare not significant factors. Multivariate analysis come to that NCAPH geneexpression, TNM stage, tumor size, and aspartate aminotransferase can affectthe disease-free survival rates of HCC patients.Conclusions:NCAPH gene may play an important role in the occurrence ofHCC development and process; if the patient preoperative NCAPH was negativeexpression, TNM stage was I-II,the tumor diameter <5cm, the AST levels <39U/L, the patient may get long-term survival.
