Objective:To study the effect of chronic intermittent hypoxia(CIH) on the expression of Bcl-2and Bax proteins in substantia nigra of paraquat(PQ) parkinson’s disease model mouse. Methods:Forty-four six-week-old male C57BL/6mice were randomly divided into four groups:control group, CIH group, PQ group and PQ+CIH group. The expression of Bcl-2and Bax proteins in substantia nigra of the mice were detected by S-P immunohistory chemistry staining. Results:1. No statistical significance were found in body weight of mice among of each group at the beginning and the end of the experiment(P>0.05).2. The positive cell number of Bcl-2and Bax of four groups in nigra:The positive cell number of Bcl-2in nigra of the mice were39.45±1.51、28.64±2.66、21.64±1.85、8.45±1.86in the control group, CIH group, PQ group and PQ+CIH group. The expression of Bcl-2was reduced gradually in four groups, the reduction of PQ+CIH group was most apparente in the among group(P<0.01),The positive cell number of Bax in nigra of the mice were11.91±1.70、20.73±1.49、30.09±2.98、52.82±2.86in the control group, CIH group, PQ group and PQ+CIH group, the expression of Bax was increased gradually in four groups,the increase of PQ+CIH group was most apparente in the among group(P<0.01). Conclusion:Chronic intermittent hypoxia combined with paraquat can upregulate the expression of Bax protein and downregulate the expression of Bcl-2protein of dopaminergic neuron in the nigra in C57BL/6mice. |
