Objective:To test the mRNA of ALDOA and PDI proteins as well as the expressionand significance of the protein in normal gastric mucosa, para-carcinoma tissue andgastric cancer tissue, so as to discuss the relationship between ALDOA and PDIproteins with the occurrence and development of gastric cancer, and to providecertain experimental data for diagnosis of gastric cancer and for the prognosisevaluation.Method: Our cases of fresh gastric cancer tissues were tested with the technologiesof Western-blot and RT-PCR, to compare with the expressions of ALDOA and PDIproteins in tissues from the cancer and in normal gastric mucosa tissues from thecancer. Then immunohistochemistry staining method was used to further observe theALDOA and PDI expressions in normal gastric mucosa tissues, atypical hyperplasiatissues, gastric cancer tissues, lymph node metastasis carcinoma tissues, and toanalyze the clinical significance of ALDOA and PDI proteins in different gastricmucosa tissues.Results:1. Western blot results show: the expression of ALDOA protein in gastric cancermucous membrane tissues was significantly higher than that near the cancer and inthe normal gastric mucosa tissues (P<0.01),The expression of PDI protein in gastriccancer mucous membrane tissues was significantly higher than that near the cancerand in the normal gastric mucosa tissues (P<0.01),2Test results of gastric cancer tissue with RT-PCR method show:The expression ofALDOA protein in gastric mucosa cancer tissues was higher than that near the cancerand in the normal tissues(P<0.01), The expression of PDI in gastric cancer tissue wassignificantly higher that in the normal gastric mucosa tissues and tissues near thecancer(P<0.01)。3. Immunohistochemistry staining results show: The Positive exPressions ofALDOA Protein in normal gastric mucosa, atyPical hyPerPlasia, gastric cancer andlymPhatic metastasis cancer tissues were resPectively8.33%(1/12),15.38%(2/13), 78.69%(48/61) and86.49%(32/37), and its Positive exPression rates in highly,medium and Poorly differentiated adenocarcinoma were resPectively36.36%(4/11),77.78%(14/18) and93.75%(30/32); the Positive exPressions of PDI Protein innormal gastric mucosa, atyPical hyPerPlasia, gastric cancer and lymPhatic metastasiscancer tissues were resPectively16.67%(2/12),23.08%(3/13),68.45%(42/61)and72.97%(27/37), and its Positive exPression rates in highly, medium and Poorlydifferentiated adenocarcinoma were resPectively36.36%(4/11),61.11%(11/18) and84.38%(27/32);4. The positive expressions of ALDOA and PDI proteins in gastric cancer were higherthan those in the normal gastric mucosa and atypical hyperplasia tissues (P<0.01);The expressions of ALDOA and PDI proteins in gastric cancer tissues were relatedwith the differentiated degrees of the tissues, which is, the lower the tissuedifferentiation is, the higher their expression are (P<0.01).Conclusion:1. The exPressions of ALDOA and PDI Proteins in gastric cancer than the normalgastric mucosa tissues increase.2. The exPressions of ALDOA and PDI Proteins were related with the occurrenceof gastric cancer, the tissue differentiated degree and the lymPhatic metastasis.
