BCCIP is a BRCA2 and CDKN1A(p21,CIP1)interacting protein,an evolutionarily conserved protein involved in maintaining genomic stability and cell cycle progression.BCCIP is widely expressed in many organizations,including skeletal muscle,heart,testis,kidney and brain,and BCCIP is a tumor suppressor associated with various tumors.Studies have shown that BCCIP deletion can significantly reduce p21 expression and accelerate the progression of G1 to S phase in L02 hepatocytes.Studies have shown that BCCIP is down-regulated in breast cancer,glioblastoma,renal cell carcinoma,ovarian cancer and colorectal cancer,but studies on the expression of BCCIP in gastric cancer have not been reported.For this reason,the expression of BCCIP protein in gastric cancer was observed for the first time.The correlation between BCCIP expression and gender,age,tumor differentiation,lymph node metastasis and tumor stage was analyzed.At the same time,the correlation between BCCIP expression in gastric cancer and Ki67 and mutant P53 was analyzed.To explore the clinical significance of BCCIP expression in gastric cancer.Objective: To detect the expression of BCCIP,Ki67 and mutant P53 in gastric cancer tissues,and to analyze the clinical significance of BCCIP expression in gastric cancer.Methods: In this study,immunohistochemical staining and Western blot were used to analyze the expression of BCCIP in 110 cases of gastric cancer and 110 matched normal gastric mucosa tissues,and then statistical analysis was used to analyze BCCIP in normal gastric mucosa and gastric cancer tissues.The difference in expression,and the correlation between BCCIP and clinical indicators of patients,including patient gender,age,tumor differentiation,lymph node metastasis,tumor stage and correlation with prognostic index Ki67 and mutant P53.Results: Immunohistochemical staining showed that BCCIP was mainly expressed in the nucleus,and 104 cases were positively expressed in 110 cases of gastric cancer.The positive expression rate was 94.55%,of which 26 cases were strongly positive,38 cases were moderately positive,and 40 cases were weakly positive.6 cases were negatively expressed,and the positive expression rate of BCCIP in normal gastric mucosa tissues matched with gastric cancer tissues was100%.There were 69 cases with strong positive expression,33 cases with moderate positive expression,8 cases with weak positive expression.68 of the 110 specimens(61.82%)showed that the expression of BCCIP in gastric cancer was lower than that of normal gastric mucosa,and the difference of BCCIP expression between gastric cancer and normal tissues was statistically significant(P<0.01).Spearman correlation analysis found that the expression of BCCIP in gastric cancer was not related to gender and age of patients,and was negatively correlated with lymph node metastasis and tumor stage(P<0.01,P<0.05),and positively correlated with tumor differentiation(P<0.01).The expression of BCCIP in gastric cancer was negatively correlated with Ki67 and mutant P53(P<0.05,P<0.01).Western blot analysis showed that the expression of BCCIP protein in gastric cancer was significantly lower than that in normal gastric mucosa(P<0.01).Conclusion:The expression of BCCIP was down-regulated in gastric cancer tissues,and negatively correlated with lymph node metastasis,tumor stage,Ki67 and mutant P53 expression,and positively correlated with tumor differentiation,suggesting that BCCIP may become a new prognostic marker for gastric cancer and may have certain significance to guide the treatment after operation.
