Objective: The research of CD4~+T lymphocyte subgroup in bacterial diarrheadiseases seldom reported.It mimic the mainly channels (infect through the mouth) inhuman gastrointestinal infection to make human infectious diarrhea disease animalmodel by gavage Enteropathogenic E coli (EPEC E2348/69). To explore the immunereaction in mice onset, it studies the level of Th1, Th2, Th17Lymphocyte cytokines inperipheral blood plasma.Method:40Healthy Kunming mouse, were randomly divided into control group (20)and the experiment group (20). The model of mouse gastrointestinal infection wasprepared with EPEC-E2348/69suspension liquid administration by gavage. The otheris normal saline control group.8hours after gavage, the observation to miceˊs shitcharacter, mental state and action was taken. Calculate diarrhea rate, diarrhea indexand the detection rate EPEC in cultivation of feces. Blood cell analysis wererespectively carried out by the collection of mice peripheral blood at the time24hbefore and after gavage.24h after bacterial gavage, the experiment and control groupsperipheral blood was taken after centrifugation. By using Cytometric Beads Array(CBA) technology, IL-2, TNF, IFN-gamma, IL-6, IL-10, IL-4and IL-17A lymphocytecytokine level in plasma was detected.Result:1. WBC(7.45±1.86)×10~9became lower after gavage WBC(6.22±1.42)×10~9and there was statistically significant difference (P=0.013<0.05); N(%) and L(%) in mice aftergavage, there was no statistically significant difference(P=0.071,0.076>0.05).Diarrheaindex of experiment group (1.12±0.66) was higher than that of control group (0.00±0.00), there was statistical differences (P <0.05), Diarrhea rate (85%) and feces cultureEPEC detection rate (75%) of experimental group were higher than that of controlgroup(0%);8hours after gavage, mice’ mental state was depressed, they moved slowly,their shit was thin, partial mice were with soft stool in experiment group; Control groupmice’ shit character and mental state had no obvious changes.2. Lymphocyte cytokine IL-2(2.55±0.32)pg/ml、IL-6(16.71±19.64)pg/ml、IL-10(12.11±3.61)pg/ml、IL-4(3.10±0.62)pg/ml、IL-17A(3.04±0.58)pg/ml in theexperiment group was higher than the control groupˊs IL-2(2.25±0.31)pg/ml、IL-6(5.96±6.75)pg/ml、IL-10(12.11±3.61)pg/ml、IL-4(2.54±0.24)pg/ml、 IL-17A(2.62±0.39)pg/ml, there was statistically significant difference (P <0.05).TNF(11.89±3.12)pg/ml、IFN-γ(25.45±69.20)pg/ml in the experiment group was higherthan the control groupˊs TNF(10.42±3.35)pg/ml、IFN-γ(7.45±7.80)pg/ml, therewas no statistical difference.3. Th1cytokines total level (39.91±71.00pg/ml) in experiment group was higher thanthat of the control group (20.13±10.46) pg/ml, there was no statistically significantdifference (P=0.237>0.05). Th2cytokines total level (34.84±20.06) pg/ml washigher than the control group (20.62±8.37) pg/ml, there was statistically significantdifference (P=0.007<0.05).Ratio of experiment group Th1/Th2(0.975) was lowerthan that of the control group (1.024), there was no statistically significant difference(P=0.832>0.05). Conclusion:1. It mimic the mainly induced channels (infect through the mouth) in humangastrointestinal infection to make human infectious diarrhea disease animal model. Thehuman infectious diarrhea disease animal model is successful.2. After mouse infected by EPEC,the level of Th1, Th2, Th17Lymphocyte cytokinesin peripheral blood plasma rises. Th1/Th2/Th17cells involved in the immune response,inflammation reaction in mice is a whole and dynamic equilibrium process.3. After mouse infected by EPEC in this experiment,Th2cells mediated the mainlyimmune response process in mice-humoral immunity, Th1cell mediatednon-dominant response.4. In animal models of EPEC gastrointestinal infection, level of Th2, Th17cytokines inplasma increase more obviously, the decrease of that of Th1/Th2hints EPECgastrointestinal infection may relative to the happening of chronic autoimmune andallergic disease.
