Objective:Effects and mechanism of panax notoginseng saponins on proliferation, apoptosis, Invasion and metastasis of hepatocarcinoma cell line HCCLM6were studied so as to illustrate the anti-tumor mechanism ofpanax notoginseng saponins, and further theoretical evidence of which as an anti-tumor drug may be provided.Methods:1. HCCLM6cells dealt with different concentration panax notoginseng saponins were then analyzed by MTT assay to calculate the cells’survival rate.2. Apoptosis rate of HCCLM6cells induced by panax notoginseng saponins were detected by Flowcytometry.3. The expression of E-Cadherin and VEGF in HCCLM6cells dealt with panax notoginseng saponins was detected by Western blot4. The change of HCCLM6cells’ adhesion force was analyzed by tumor adhesion test.5. The change of HCCLM6cells’s motility and invasion ability were analyzed by Transwell assay.6.Statistics Methods:Statistic analysis was performed with SPSS13.0. All data in this experiment were indicated Mean±STD.Results:1. Panax notoginseng saponins inhibites the proliferation of HCCLM6cells. By MTT analysis, it is shown that proliferation of HCCLM6cells was significantly inhibited by panax notoginseng saponins at concentrations of25、50、100、200、400mg/L (P<0.05), and showing a dose-and time-dependent manner from24h to48h.2. Panax notoginseng saponins can induce HCCLM6cell apoptosis.After treatment with different concentration (25、50、100、200、400mg/L) panax notoginseng saponins for48h, apoptosis rate of HCCLM6cell compared with the grupe dealed with nothing is rised. After incubated with200mg/L panax notoginseng saponins for48h, the apoptosis rate of HCCLM6cells increased compared with uncontrol group (P<0.01).3. Panax notoginseng saponins can rises expression of protein E-cadherin and lessens espression of VEGF in HCCLM6cells.4. Panax notoginseng saponins can reduce the motility and invasion of HCCLM6cells.After dispose of panax notoginseng saponins (200mg/L), the number of HCCLM6cells that through the artificial basement membrances had a significantly reduction. The inhibitory rate of HCCLM6cells arrived to%(P<0.001). By MTT analysis, it is shown that the adhesion ability of HCCLM6cells was significantly inhibited by panax notoginseng saponins (200mg/L), the inhibitory rate of HCCLM6cells increased from100%of control group to%(P<0.00). After treatment with panax notoginseng saponins, the protein expression of E-cadherin inHCCLM6cells showed a bemarkedlyincreased and the protein expression of VEGF is lessend by Western blot.Conclusions:Panax notoginseng saponins can inhibit proliferation, induce apoptosis, and reduce hepatocarcinoma cells adhesion, invasion and motility ability. The panax notoginseng saponins resistance mechanism of invasion and metastasis in tumor may be related to the increase of the protein E-cadherin and the decrease of VEGF.Panax notoginseng saponins might be a potential of liver cancer drugs.
