Background and Objective:Congenital heart disease (CHD) is a fetus in the womb, the heart and great vessels developmental abnormalities caused by congenital malformations. Ventricular septal defect (VSD), atrial septal defect (ASD), patent ductus arteriosus (PDA) and so on with left to right shunt congenital heart disease is often secondary to systemic circulation-pulmonary circulation shunt pulmonary arterial hypertension. the study of animal pulmonary arterial hypertension induced by artificial shunt is important means that we realize systemic circulation-pulmonary circulation shunt pulmonary arterial hypertension secondary to congenital heart disease. In study, the use of the improved "casing" method establishing rat carotid artery-venous shunt model to observe patency situation of shunt and pulmonary arterial hypertension Whether there is.Research Methods:40SD rats of body weight270-290g were divided into shunt group (n=29) and control group (n=11). Shunt group are establish rat common carotid artery and external carotid vein of left to right shunt with a long about5mm polyethylene casing. Control group only ligate rats left common carotid artery and external carotid vein, don’t build rat common carotid artery and external carotid vein of left to right shunt.(1) to observe Shunt group patency situation of carotid artery-vein shunt immediately after surgery and3months after surgery.(2) to observed in shunt rats whether changes in pulmonary arterial hypertension:After3months, compared with two groups of rat, whether there are differences in Mean pulmonary artery pressure (mPAP), pulmonary vascular remodeling index(WT%and WA%), right ventricular hypertrophy index.Results:(1) completion of the surgery16postsurgical instantly are seen clearly in the carotid-venous shunt, the immediate postoperative patency rate of carotid artery-vein shunt was100%,3months after surgery, surgical exploration found that all Shunt group rats carotid-venous shunt is blocked, the rat left common carotid artery and left external carotid vein disappears, polyethylene casing is wrapped in fibrous tissue.3months after surgery, the postoperative patency rate of carotid artery-vein shunt was0.(2) pulmonary arterial hypertension observed indicators:mPAP of shunt group compared with mPAP of control group no difference (P>0.05). Rat pulmonary artery was observed under an optical microscop that Shunt group rats did not appear typical pathological changes of pulmonary hypertension(vascular intimal thickening, smooth muscle hyperplasia, thrombosis, stenosis and occlusion, etc). Two groups of rats pulmonary vascular remodeling index (WT%and WA%)does not exist differences (P>0.05). The shunt group rat right ventricular wall thickness compared with the control group no significant thickening, the Shunt group rat of right ventricular hypertrophy index have no obvious rise (P>0.05).Conclusion:1. Rat carotid artery-vein shunt model was successfully established.2. Invasive testing of rat pulmonary artery pressure is completed.3. Detection of Experimental design is completed.4. Early occlusion of rat carotid artery-vein shunt is the bottleneck of further research.
