Objective:To construct mouse model of acute myeloid leukemia and detect the expression of ANGPT1,ANGPT2and VEGF gene, as well as the clinical significance.Methods:1. HL-60cells were transfected with NOD/SCID mouse through abdominal injection to construct mouse model of acute myeloid leukemia.2. To identify mouse model by histopathology and Flow Cytometry.3. The expression of ANGPT2, ANGPT1and VEGF mRNA in the tumor tissues of mouse model was detected by real-time fluorescence quantitative PCR.4. The expression of ANGPT2will be analysed on the relationship with the survival time of mouse model by spearman’s correlation method.Results:1. Mouse model has been successfully identified by histopathology and Flow Cytometry.2. The expression of ANGPT2and VEGF in mouse mode was significantly detected higher than normal group (P<0.05).But the expression of ANGPT1was lower than ANGPT2and VEGF, there was no significant difference in ANGPT1and normol group (P>0.05)3. The increasing expression of ANGPT-2in mouse model had a short survival time in acute myeloid leukemia.Conclusions: This study showed that ANGPT-2mRNA was overexpressed in acute myeloid leukemia. The increasing expression of ANGPT-2mRNA may lead to poor prognosis in acute myeloid leukemia. More importantly, it provided basis knowledge and experiment for the ANGPT-2target therapy for acute myeloid leukemia.
