[Objective]:By detecting the relationship of EZH2、P53protein expression between protein expression in normal colorectal tissues and colorectal cancer tissues and tumor various clinical pathological parameters and expression of two proteins, to further explore the molecular biological mechanisms of the change from normal colorectal tissue to colorectal cancer and find new molecular markers for the knot to the taxation and targeted gene therapy of cancer biological characteristics.[Methods]:Immunohistochemistry was used to detect the expression between EZH2and P53gene proteins in40cases of colorectal cancer and20cases of normal colorectal rectal mucosa organization, a total of60cases of paraffin-embedded tissue, and then SPSS17.0statistical package, the chi-square test and Spearman rank correlation analysis method were used to analyze the datas.[Results]:1The expression of protein EZH2and P53protein was mainly localized in the nucleus, cytoplasm as well as some expression.2The positive expression rate (30.00%) of EZH2protein in colorectal carcinoma was higher than that (5.00%) in normal colorectal tissue, and there was significant difference between the two groups(P<0.05). There was the statistically significant(P<0.05)among EZH2expression and the lymph node metastasis、differentiation and TNM stage of the colorectal cancer. The positive expression rate:the lymph node metastasis group (50.00%) was higher than without lymph node metastasis (13.64%); the poorly differentiated group (62.50%) was higher than that (33.33%), high (15.00%) differentiated group; the clinical stage III-IV period (47.62%) was higher than in stage Ⅰ~Ⅱ (10.53%). There has not yet found the statistically significant (P>0.05) with gender, age, tumor location, tumor size, pathological type and preoperative CEA.3The positive expression rate (77.50%) of P53protein in colorectal carcinoma was higher than that (30.00%) in normal colorectal tissue, and there was significant difference between the two groups(P<0.05). There was the statistically significant(P<0.05)among P53expression and the lymph node metastasis, differentiation and TNM stage of the colorectal cancer. The positive expression rate:the lymph node metastasis group (94.44%) was higher than without lymph node metastasis (63.64%); the poorly differentiated group (100.00%) was higher than that (91.67%), high (60.00%) differentiated group; the clinical stage III-IV period (90.48%) was higher than in stage Ⅰ~Ⅱ (63.16%). There has not yet found the statistically significant (P>0.05) with gender、age、tumor location、 tumor size、pathological type and preoperative CEA.4EZH2and P53protein expression in colorectal cancer was positively correlated (rs=0.748, P<0.05)[Conclusion]:1EZH2protein expression in colorectal cancer was higher than the level of expression in normal colorectal tissue, and there were significant differences (P<0.05), which suggestted that EZH2protein might play a role in promoting the development from normal colorectal tissue to colorectal cancer. EZH2protein high expression and lymph node metastasis increases, degree of differentiation deteriorate, and invasion infiltration deepening of colorectal cancer were positively correlated. This prompted that EZH2protein, as the expression product of the EZH2candidate oncogene, could promote colorectal cancer cell invasion and metastasis.2P53protein mutant type expression in colorectal cancer was higher than the level of expression in normal colorectal tissue, and there were significant differences(P<0.05), which suggestted that P53protein mutant type might play a role in suppressing the development from normal colorectal tissue to colorectal cancer. P53protein mutant type high expression and lymph node metastasis increases, degree of differentiation deteriorate, and invasion infiltration deepening of colorectal cancer were positively correlated. This prompted that P53protein mutant type, as a cancer-promoting protein, can promote colorectal cancer cell invasion and metastasis.3EZH2and P53protein mutant type expression was positively correlated in colorectal cancer.Two proteins in colorectal cancer evolution process in addition to their own direct role, P53protein is mutated, the loss of silence EZH2protein mechanism that may cause increased EZH2expressionand and participate in colorectal cancer cell proliferation, dedifferentiation and invasion increases and metastasis increases.4Colorectal carcinoma P53gene expression similar to other tumor tissues, suggesting that P53gene can also be used as a possible gene therapy approaches in colorectal cancer. But need to further explore and research.
