Objective:Observe the functional and moophologic changes after the rats’ kidneys accepted radiation, To investigate the impact of single electronic ray irradiation on renal damage in rats so as to provide a reference for its clinical application.Methods:A total of120SD rats were equally randomized to five groups as control,8Gy-,10Gy-,12Gy-and15Gy-group,every group contains24rats.The rats were anesthetized with10%chloral hydrate(0.3ml/100g), proned on the specially crafted mold, with rats’four limbs stretching.placed them in the simulator machine to define the kidney’s position.Put the rats in the linear accelerator(siemens Primus M), The limited light tube limited the radiation field2.5cm×2.5cm aera. radiation doses were8Gy,10Gy,12Gy and15Gy. The dose rate was300cGy/min,source skin distance was100cm. Control group pretended accepting irradiation after anesthesia.Renal functions were examined at the1st,3rd,6th,9th month after radiation. Morphological changes were observed by HE staining.Results:Compared to control group,renal index of≥8Gy groups significantly reduced in9th month;the systolic pressure of group15Gy was increased significantly in9th month.Scr and BUN of≥12Gy-group increased significantly since6th month, group8Gy and10Gy were increased significantly in9th month. UP/UC of group15Gy was significantly increased since3rd month,while in other groups were significantly increased since6th month.Radiation more than lOGy can up-regulate the CysC significantly from6th month,while the8Gy from9th month.In all irradiation groups,reduction of renal volume,decrease of proportion of cortex can be seen.There are pronounced mesangial dissolution,then exhibit mesangial sclerosis. Thickening of capillary loops,dilatation of lumens of blood vessels,and increasing of renal tubule cell spaces could be seen.Part of the epithelial cell swelling,dissolution, necrosis.Conclusion:The single radiation from8to15Gy may cause radiation damage in the following9months to rat kidney, being dose and time-dependent.
