Objective:Esophageal carcinoma is the most cause of cancer deaths world wide. Only10-20%of all esophageal carcinoma patients are alive5years or more after diagnosis. More and more medical research focus on looking for gene markers which correlate with esophageal cancer and could be used for early diagnosis and judgment of prognosis, risk of recurrence and metastasis, even for target of genetic therapy.PLK1(polo-like kinase1) is considered to closely relate with mitosis. Recently many researches revealed that the over-expression of PLK1(or PLK) were found in various primary carcinoma, and indicated the relationship between PLK1and human cancers biological feature and prognosis are very tightly.The purpose of this study was to determine whether PLK1is over-expressed in surgical specimens of esophageal carcinoma as compared with normal esophageal tissue and to assess its relation to pathological classification TNM staging and metastasis by immunohistochemical method. Methods:In the present study, immunohistochemical method was performed to detect PLK1and P53in63esophagealc carcinomas as well as10normal esophageal specimens. Pathological TNM staging was performed according to AJCC2009protocol. SPSS17.0for Windows Software was used to analyze Statistical data.ResultsPositive staining for PLK1was observed in73.0%of the esophageal carcinoma specimens, and negatively in normal esophageal specimens. The expression level elevation in the malignant group was statistically significant compared with that of the normal group.There were no significant associations between PLK1and age.and grade. PLK1expression was significantly related to histological type, tumor position,lymph node metastasis and TNM staging. In esophageal carcinoma P53expression was significantly associated with PLK1.Conclusion:1. the over-expressed PLK1in esophageal carcinoma specimens, compared with that of normal esophageal specimens, indicated that PLK1is a new tumor marker for esophageal carcinoma.2. PLK1expression level in esophageal carcinoma have correlation with the histological type, tumor position,lymph node metastasis, and TNM staging. It indicated that PLK1might be a prognostic marker. 3、 No statistical significance was found between the PLK1expressions of different ages and grades.4、 Statistical relation was found between the PLK1expression level and the accumulation of P53, which indicated P53might be one of the targets of PLKl.
