Objective:To investigate the proteome analysis of sperm in different AID outcomes using two-dimensional gel electrophoresis (2-DE) coupled with mass spectrometry (MS), and to discuss the mechanism of different outcomes.Methods:1、Sperm from one donor that failed in9continuous AID cycles was attributed as experimental group, and sperm from another three donors that succeeded within50%AID cycles was assigned as control group.2、 Collecting the sperm and extracting all of the sperm proteins3、Analysing the differences between the two groups of sperm proteins using two-dimensional gel electrophoresis (2-DE), coupled with mass spectrometry (MS)4、Western-blot were employed to explore the expression pattern of Semenogelin-1and Glutathione S-transferase Mu3which are the identified proteins between the two groups using2-DE coupled with MS.Results:1.There were432protein spots in experimental group and460protein spots in control group in the showing gel map. Three hundred forty seven protein spots in experimental group could match up with those in control group. Twenty two different protein spots were detected, among which17protein spots were detected by MS2.Semenogelin-1and Glutathione S-transferase Mu3were highly expressed in the experiment group.Conclusions:The difference proteins of one donor’s sperm suffered continuous AID failures were analyzed by2-DE coupling with MS and found22difference proteins. Thereafter, the expression change of Semenogelin-1and Glutathione S-transferase Mu3were validated by using Western-blot. However, its biological significance needs to be further investigated.
