Objective A study was conducted to compare the difference between idiopathic oligoasthenoteratospermia and varicocele with oligoastheno-teratospermia on sperm protein, and evaluate the significance of the differences in sperm protein.Method Patients who were diagnosed with idiopathic oligoastheno-teratospermia and varicocele with oligoasthenoteratospermia in the Guangxi Zhuang Autonomous Region people’s Hospital from January to December in 2013 were included. Varicocele with oligoasthenoteratospermia and idiopathic oligoasthenoteratospermia were in the experimental group (n=32) and control group (n=32) respectively, and the semen specimen were collected. Urea, guanidine hydrochloride and supersonic schizolysis method were used to extract protein. Two-dimensional gel patterns were obtained by using two-dimensional gel electrophoresis, staining, fixing, rinsing and scanning methods. PDQuest differential protein analysis software was used to compare the differences in protein and gel point. The last step was protein identification via mass spectrometry. Statistical software spss16.0 was used to analyze data.Result:DQuest software analysis showed that the detection total protein points in the experimental group and the control group were 759 and 661, respectively. According to the expression quantity of 2 times to calculate, high expression protein in the experimental group and the control group were 17 points and 11 points respectively. While according to 5 times calculate, there were 6 points high expression in the experimental group and 3 points in the control group. Form the gel could dig out one most differential protein spot in each group, respectively. Protein identification via mass spectrometry and protein date base search showed that the protein high expression in the experimental group and the control group were Tubulin beta-4B and Lactotransferrin, respectively.Conclusion By protein electrophoresis and mass spectrometry analysis, this study showed that there are differences between two groups of patients in sperm protein and found out two differential proteins. The lack of the two differential proteins may have an impact on the complex life of sperm and infertility, could be one of the cause of infertility that associated with idiopathic oligoasthenoteratospermia and varicocele.
