Objective1. Production of heparin selective sheep acellular dermal matrix (acelluar Dermal Matrix ADM).2. Use of the heparin sheep ADM treatment of rats with deep second degree burns to study burn wound healing rates and wound tissue changes in the structure.3.Explore heparinized sheep ADM treatment burn wounds, wound tissue changes of apoptosis, which reveals the biological dressings in promoting wound healing mechanisms. Method1-year-old healthy sheep to take thickness skin produced selective sheep ADM.2.160healthy Wistar rats were made into a deep second degree burns were randomly divided into iodophor gauze treatment group, pigs ADM treatment group, sheep ADM treatment group and heparin sheep ADM treatment group. Scalded rats during surgery3d,7d,14d,21d and28d calculate the rate of wound healing in rats. The rats were killed and at the same time in each time period, to take the wound tissue specimens HE staining and in situ TUNEL apoptosis staining, wound healing was observed in different periods within the organizational structure change. Results1. Successfully fabricated selective sheep ADM, epidermal integrity, no cellular components, blood vessels and skin appendages.2Fourteen days after burn,21d,28d, pig ADM group, group of sheep ADM and heparinized sheep ADM group wound healing rate than povidone-iodine gauze group (P<0.05),21d heparinized sheep ADM group better than pig ADM group (P<0.05),21d and28d heparinized sheep ADM group than sheep ADM group (P<0.05).3groups wound HE staining21d, pig ADM group and sheep the ADM group dermis layer arrangement of collagen fibers after injury less structured, fibroblasts began to appear more growth. Heparinized sheep ADM group after injury21days epidermis, dermis structure the basic comparison rules of normal arrangement of collagen fibers.5groups of deep second degree burns after at7d, groups wound tissue fibroblast apoptosis were significantly increased compared to3d, but each group, see no significant statistical difference (P>0.05). With rat wound healing, in burns after14d-28d, the wound tissue in the number of apoptotic fibroblasts significantly increased apoptosis in pig ADM group, sheep the ADM groups and heparinized sheep ADM group number compared with povidone-iodine gauze group increased significantly (P<0.05), especially heparinized sheep ADM treatment group in the the postoperative28d digital highest (P<0.05). Conclusion1. Successfully produced selective sheepskin dermal matrix.2. Three dermal matrix treatment of deep second degree burn wounds, especially in heparinized sheep ADM better promote wound healing organizational structure recovery effect.3. Heparinized sheep ADM in burn wound healing post, more than other groups to induce or promote apoptosis of granulation tissue fibroblasts, reduce scar formation, so as to promote wound healing.
