Objective: The aim of the study was to analyze the pathogenicity of candidaalbicans through Candida albicans adhere to KB cell, compare the expression ofinterleukin-6of KB cells under the stimulation of Candida and without stimulation,toinvestigate the possible effection of oral epithelial cells defense candida infection.Methods: Oral squamous cell carcinoma cell lines KB cell as the research object,the KB cell infected by Candida albicans after different time, quantify the number ofCandida albicans cells adhering to the KB cell, observed the Candida albicans viabilityand determine the transformation of Candida albicans from blastosporess to hyphae.Coculture KB cell with blastospores phase,hypha phase and inactivated Candidaalbicans for6,12,24hours and KB cells were cultured alone as the control group. At theend of each contact period,the culture supernatants were harvested, IL-6protein levelswere analyzed by enzyme-linked immunosorbent assays (ELISA). Determine theexpression of IL-6mRNA during KB cells infected by Candida albicans by reversetranscription polymerase chain reaction (RT-PCR).Results:1. Candida albicans begin adhere to KB cell after it was infected, thenumber of KB cells adhered was increased gradually within24hours of infection andblastospores phase and hyphae phase Candida albicans all can adhere to KB cells.2.Viability and growth analyses showed that all Candida albicans that adhered to KBcells were alive, however, the sum of free candida were reduced as time extend from1.5×105/cm~2after a3h infection to around8×103/cm~2after a24hours infection.3.Thetransformation of candida from blastosporess to hyphae was time dependent. Thepercentage of the hyphal form rose as time extend from23.1%after a3h contact tomore than82.2%after a24hours contact. The difference was statistically significant,P<0.05.4.The level of IL-6peptides of experiment group was higher than the control group.The expression of IL-6peptides was time dependent,achieve a peak at12hoursand then appeared downturn. IL-6peptides expression of the blastospores group andthe hypha group were higher than the control group,P <0.05, and the difference wasstatistically significant.5.KB cell was coculture with blastospores phase and hyphaephase Candida albicans for24hours,the expression of IL-6mRNA was higher thancontrol group, P <0.05, and the difference was statistically significant.Conclusion:1.The number of KB cells adhered and the sum of free candida weretime dependent through Candida albicans adhere to KB cell. The transformation ofcandida from blastosporess to hyphae and time were positively correlated.2. Candidaalbicans to KB cells induced expression of IL-6peptides,and the expression of contentwas time dependent.3.Candida albicans to KB cells induced expression of IL-6mRNA.Candida albicans is responsible for the production of IL-6secreted by epithelial cellswhich can be expected to play an important role in the immune response.
