Objective: To investigate the effect of dioscin on TNF-α induced inflammation inHUVECs.Methods:HUVECs’ injury model induced by TNF-α was established and cellswere divided into five groups, three of which were interfered with different does(0.3125μg/ml、0.625μg/ml、1.25μg/ml) of dioscin. MTT method was used to detect cellvigor while RT-PCR was used to detect VCAM-1mRNA expression of each group.Adhesion of monocytes to HUVECs was observed by microscope, and the function ofU937derived macrophage-swallow-DiI-oxLDL was observed by fluorescencemicroscope.Results: Compared with control group, the cell proliferation in TNF-α group wasobviously increased (P<0.05). After incubation with dioscin, the proliferation wassignificantly reduced than that in TNF-α group(P<0.05). TNF-α group couldsignificantly increased VCAM-1mRNA expression (P<0.05) and this effect could beinhibited greatly by dioscin in a dose-dependent manner(P<0.05). Also dioscin couldinhibit U937adhesion to HUVECs, and finally BBR attenuated OX-LDL uptake bymacrophages.Conclusion: Dioscin could inhibit the inflammation of HUVECs induced byTNF-α.
