Objective: This experiment is to build a model of dogs’ chronic apicalperiodontitis by putting Agnanowith different consistency into their root canals in orderto make a research on its bacteriostasis effect.Method: This experiment chose three adult dogs and took the teeth of I3, P3andP4on the right side of maxilla as research objects. The pulp chambers of the three teethwere opened, the pulp was extirpated and the teeth were exposed in oral cavity for1w.After that, glass ionomer cement was used to seal up the holes of the pulp chambers for6w, then X-rays of them were taken to see the condition of dogs’chronic apicalperiodontitis. According to different dental positions, the three dogs were classified intothe group of1%Agnano, the group of2%Agnanoand negative control group, and thecorresponding experimental medicine was put into their root canals. After4w, X-rayswere taken, the medicine was fixed with the method of internal infusion and the teeth,along with alveolar bone were used to make Talor’s improved tissue section under theapplication of Gram’s staining. The work of bacteria counting was done and finally, theexperimental data was statistically analyzed with SPSS.Results:(1) Results of Researching the X-raysAfter the period of6w of the seal of pulp chambers’ holes with glass ionomercement, the experimental teeths’periapical alveolar bones were obviously absorbed, andthe apical shadow area is approximately1.5~2mm2.After the4w’s seal of the experimental medicine in Root-Canal, the apical shadowarea of1%Agnanosolution group became obviously small (about0.5mm2); that of2%Agnanosolution group disappeared; and that of negative control group did not obviously change.(2) Results of Researching Talor’s Improved Tissue Section under the Applicationof Gram’s StainingIn the negative control group, large bacteria coverage with deep blue staining canbe seen; in1%Agnanosolution group, a medium number of bacteria can be observed; in2%Agnanosolution group, a few can be seen. The number of bacteria in the negativecontrol group is larger than that in1%Agnanosolution group and2%Agnanosolutiongroup, and the number of bacteria in1%Agnanosolution group is larger than that in2%Agnanosolution group. The above difference all has statistical significance (P□0.05).Conclusions:1%and2%Agnano-PLGA both have effectively bacteriostasis effect of dogs’chronic apical periodontitis,and the bacteriostasis effect of2%Agnanois better than1%Agnano.
