Objective:To discuss and compare the anti-tumor effects of celecoxib andletrozole on breast cancer cell MCF-7through investigating the effects of celecoxib andletrozole, respectively, and combined, on the growth of human breast cancer cellMCF-7.Methods:1.Investigate the effects of different concentrations of celecoxib (0,20,40,60,80and100umol/L),and different concentrations of letrozole(0,100,200,400,600, and800umol/L), respectively, and combined(celecoxib40umol/L+letrozole200umol/L),through MTT assay, compare the proliferation and inhibition rate of human breastcancer cell MCF-7transfected by COX-2,after72hours of drug delivery. Drug-freeculture medium as the negative control group.2.Investigate the effects of different concentrations of celecoxib (60,100μmol.L-1),and different concentrations of letrozole(200,400μmol.L-1), respectively,and combined(celecoxib40umol/L+letrozole200umol/L), through cell migratoryinfiltration assay, compare the proliferation and inhibition rate of human breast cancercell MCF-7transfected by COX-2,after72hours of drug delivery.Drug-free culturemedium as a negative control group.Results:1.The proliferation and inhibition rate of different dose groups of celecoxib onMCF-7cell increase with increasing drug concentration (P <0.01).2.The proliferation and inhibition rate of different dose groups of letrozole onMCF-7cell increase with increasing drug concentration (P <0.01).3.The survival rates of MCF-7cells were53.8%and68.3%,with celecoxib(40umol/L) and letrozole (200umol/L)alone after72hours of drug delivery.4.The survival rates of MCF-7cells were36.6%,with celebrex40umol/L+ letrozole200umol,lower than the monotherapy group.Conclusion:1.Celecoxib and letrozole both can inhibit the proliferation and migration of breastcancer cell MCF-7,with dose-dependent.2.Celecoxib and letrozole combined have collaborative effects on anti-breastcancer, with dose-dependent.
