Objective:To study the relationship of hormonal level and clinic effect of letrozole as endocrine therapy for postmenopause metastatic breast cancer.Methods:27 patients were assigned to receive letrozole 2.5mg,po,qd.Plasma estradiol concentrations were determined by RIA before treatment and 8,16 week after treatment.Clinic effect and estradiol level were evaluated.Result:Objective response rate was 46.2%.The effective rate of E2(> 90pmol/L) was 26.7%, The effective rate of E2(20-90pmol/L) was 72.7%.The effective rate of menopause time(<5years) was 25%,The effective rate of menopause time(≥5years) was 55.6%.The effective rate of different receptor(ER and PR+) was 66.7%, The effective rate of different receptor(ER or PR+) was 28.6%.The effective rate of Her-2(+) was 25%,The effective rate of Her-2(-) was 64.3%.The effective rate of different metastasis genetic(internal organs) was 20%,The effective rate of different metastasis genetic(outward organs) was 62.5%.Plasma level of estradiol was supperessed by 41%.The major drug related adverse events were nausea and angina,debilitation. Concl-usion:Letrozole is effective and safe as hormonal therapy for postmenopausal metastatic breast cancer.
