Objective:This study is to explore the correlation between polymorphism in-3148C/G site of GDF-15gene and the Acute Myocardial Infarction (AMI) associated with the formation of collateral circulation in Taiyuan Han nationality.Methods:Use the case-control study, collected the92AMI patients (within3-12hours) as case group, of which68patients in a collateral group,24patients in a non-collateral group and56normol subjects as a control group. Both groups were collected from Taiyuan Han population. DNA was isolated from peripheral blood, use the Restriction Fragment Length Polymorphism polymerase chain reaction(RFLP-PCR)、gene sequencing and sequence flanking to analysis the correlation of GDF-15gene-3148C/G with the Acute Myocardial Infarction (AMI) with formation of collateral circulation.Results:There are2genotypes in the GDF-15gene-3148C/G polymorphism in Taiyuan Han population:CC and GC. The GG genotype was not detected. The AMI group and control group are suit for Hardy-Weinberg law, have group representativeness The genotype frequencies of CC、GC were80.43and19.57%in the AMI group and in the control group were60.71%and39.29%respectively. Testers of the two groups at-3148C/G CC, GC genotype frequencies distribution have a statistics mean (X2=6.864, P=0.009);The risk genotype was GC, Compare to the CC genotype, GC genotype increase the risk of AMI for2.6folds.(OR=2.660,95%of confident interval is1.265-5.595). And the genotype frequencies of CC-. GC were85.29%and14.71%in the AMI collateral group and66.67%and33.33%in the AMI non-collateral group individually, which the testers of two groups at-3148C/G CC, GC genotype frequencies distribution have a statistic significance.(X2=3.911, P=0.048); The risk genotype was GC, Compare to the CC genotype, GC genotype increase the degree of formation of collateral circulation for2.9folds (OR=2.900,95%of confidence interval is0.983-8.556).Conclusions:People with GC genotype at GDF-15gene-3148C/G site may have a higher risk for the occurrence of AMI. And people with GC genotype more contribute to the formation of collateral circulation. Detect the GDF-15genotype maybe as the biomarker of early diagnosis and therapy.
