Objective:To explore the expression levels of SFRP1in tissue and serum of cervical squamous cell carcinoma (SCC), cervical intraepithelial neoplasia (CIN) and cervicitis, and analysis the effect of SFRP1in the development of cervical cancer.Method:1. Sample collection.2. HE staining analysis for the histological differences between the cervical SCC, CIN and cervicitis.3. Immunohistochemistry analysis of the expression of SFRP1in cervical SCC, CIN and cervicitis tissues.4. Real-time PCR quantitative analysis for tissue expression of SFRP1gene in cervical SCC, CIN and cervicitis.5. Western blotting analysis for protein expression of SFRP1in cervical SCC, CIN and cervicitis tissues.6. ELISA detection for SFRP1expression in serum of cervical SCC, CIN and cervicitis patients.Results:HE staining showed cervicitis had squamous columnar epithelium vascular proliferation in submucosal and mucosal tissues, dilatation and congestion, accompanied by chronic inflammatory cell infiltration. Cervical intraepithelial neoplasia (CIN) appeared nipple pseudo fiber vascular factors. Squamous cell hyperplasias were mainly acanthosis hyperplasia. Cervical carcinoma (SCC) showed layered cell proliferation. The cells were pleomorphic and had cancer cells groove. Cervical cancer tissue showed highly differentiated squamous cell carcinoma. Cervicitis shows submucosal and mucosal squamous columnar epithelium vascular proliferation, dilatation and congestion, accompanied by chronic inflammatory cell infiltration. HE staining showed that the layered cell proliferation appeared in cervical SCC tissue. Cell was pleomorphic and had complex phenomenon with increased mitosis. Epithelial cell showed layered split phenomenon.2. Immunohistochemical staining showed that SFRP1protein was mainly expressed in the cytoplasm of CIN and the cervicitis group, with brownish yellow or brown granular staining. However, there was no significant expression in cervical SCC tissue.3. Quantitative PCR analysis showed that sfrpl gene expression levels in cervical SCC and CIN group were significantly reduced compared with cervicitis group.4. The expression levels of SFRP1protein were significantly reduced in SCC compared with CIN and cervicitis by Western blotting.5. ELISA detection showed that SFRP1protein expressing in serum of cervical SCC and CIN were significantly reduced compared with cervicitis. No significant difference of SFRP1protein was found in the serum of patients between the CIN and cervical SCC.Conclusion:1. SFRP1may play an important role in the development of cervical SCC. Lower levels of SFRP1expression promoted the development of the cervical SCC.2. The expression levels of SFRP1protein significantly decreased in the patient’s serum of cervical SCC and CIN. SFRP1may be an early diagnostic indictor of cervical SCC.
