Object:In order to clarify the mechanism of three major drugs,radix rehmanniae recen, root of common peony, Oldenlandia, in the formula "detoxification removing stasis and nourishing yin" in the Prevention and treatmentof lupus nephritis and their influence on glucocorticoid receptor alpha and bone morphogenetic protein-7expression. To explore the relativeness of lupussugar rat kidney cortex hormone receptor a and bone morphogenetic protein-7expression.To elaborate the mechanisms of nourishing yin drugs in the formula for the prevention and treatment of lupus nephritis.Methods:12-week-old female MRL/1pr mice were selected as experimental subjects randomly divided into six groups, the control group(n=8), saline20ml.kg-1.d-1; glucocorticoid group (n=8). Glucocorticoid+the detoxification removing stasis and nourishing yin group8, be prednisone suspension10mg. kg-1.d-1+detoxification removing stasis and nourishing yin20g.kg-1.d-1. Glucocorticoid+dried radix rehmanniae group of eight, to be of prednisone suspensions10mg.kg-1.d-1+dried radix rehmanniae2.5g.kg-1.d-1;8the glucocorticoid hormone+red peony root group, be prednisone suspensions10mg kg-1.d-1+TPG2g.kg-1.d-1; glucocorticoid+Hedyotis diffusa group8be prednisone suspensions10mg kg-1.d-1+Hedyotis diffusa2.5g. kg-1.d-1. Every four weeks urine was collected. After8weeks, urinary protein excretion of the mice in each group, were measured, competitive inhibition of enzyme-linked immunosorbent assay determination of the rate of change of serum ds-DNA antibody binding; and the expression of.the kidney pathological examination and Western Blot, determination of renal tissue glucocorticoidestrogen receptor alpha and bone morphogenetic protein-7. Results:1.The general state of lupus mice:control group:weight gain was significantly slow, body hair fluffy, dull, less active, chilling, back arched, weak stress ability, mortality was higher; whereas in the treatment group after the drug intervention symptoms have various degrees of ease, lupus mice survival situation:a model group mice died.2.The measurement of24h urinary protein excretion of the mice:before gavage: lupus mice urine was no significant difference in the amount of protein; After gavage for4weeks, the all-party group and root of common peony group lupus mouse urinary protein content with the control group is decreased, the difference was statistically significant (P<0.01), the all-party group urinary protein levels is below root of common peony group; after8weeks, the hormone group, the all-party group and dried radix rehmanniae group, the red peony groups and Hedyotis diffusa group lupus mice urine protein was significantly lower than the control group, the difference was statistically significant (P<0.01), and urineprotein levels from low to high:red peony group<dried radix rehmanniae group<all-party group<Oldenlandia diffusa group<hormone group<control group.3.Serum ds DNA antibodies Determination:The all-party group and the hedyotis diffusa group lupus mice of ds-DNA antibody levels decreased. compared with control group, the difference was significant (P<0.01) ds a DNA antibody levels ranging from low to high:the all-party group<Hedyotis diffusa group<root of common peonygroup<dried radix rehmanniae group<hormone group<control group4.Mouse kidney tissue GR-a determination:the lupus mice kidneys of the all-party group and root of common peony group of GR-a control group significantly increased (P<0.05)。5. BMP-7was measured in the mouse kidney tissue:the all-party group and root of common peonygroup, BMP-7and the control group increased significantly (P<0.05)6. pathomorphological changes:control group presentation to severe mesangial cell proliferation, mesangial matrix significantly widened glomeruli showed focal segmental glomerular sclerosis, capillary endothelial cell hyperplasia, stenosis obvious interstitialinflammatory cell infiltration, the most serious one. hormone group lupus rat kidney mild to moderate mesangial cells, mesangial matrix with mild to moderate widened capillary endothelial cell proliferation compared with the model group reduced, part of the capillary lumen remained narrow, with interstitial inflammatory cell infiltration reduced, lesion mild improving; The all-party group and the dried radix rehmanniae group only widened mildly in mesangial cell proliferation and matrix, capillary endothelial hyperplasia is not obvious, lumen patency approximately like the normal glomerular structure. Interstitial inflammation the cell only can be seen a small amount of inflammatory cell infiltration, the lesions improved significantly.Conclusions:The formula "detoxification removing stasis and nourishing yin" and hedyotis diffusa of it Can significantly reduce the level of anti-double-stranded DNA antibodies in lupus mice; The formula "detoxification removing stasis and nourishing yin" and red peony root of it can make Lupus mice significantly reduced urinary protein levels, and can significantly improve the GR-a and BMP-7level and relieve lupus mice kidney fibrosis. GR-a in lupus mouse kidney and BMP-7have a high relevance, The formula "detoxification removing stasis and nourishing yin" and dried radix rehmanniae of it can influence the BMP-7content of their kidneys by regulating the GR-a in lupus mouse kidney to further reduce or even improve lupus mice kidney damage.
