The Prevention Of99Tc-MDP On Glucocorticoid–induced Osteoporosis Rabbit Model

Objective: To compare the calcium and alendronate with B-ALP、TRAP-5b andpathologic tissue of osteoporosis, understanding the prevention of99Tc-MDP onglucocorticoid-induced osteoporosis rabbit model.Methods: The forty healthy female New Zealand rabbits were randomly dividedinto five groups: normal control group(group A), the model of osteoporosis in thecontrol group(group B), model of osteoporosis+99Tc-MDP treatment group(group C),model of osteoporosis+alendronate treatment group(group D), model of osteoporosis+calcium carbonate D3treatment group(group E). There were eight rabbits in the eachgroup. The groups of B、C、D and E were given hydrochloric dexamethasone3mg/kg(im) twice a week for six weeks, after six weeks changed to once a week. Meanwhile,the group C was given99Tc-MDP4mg/3ml/week, through the era vein injection; thegroup D was given alendronate tablets1.82mg/kg/week; the group E was givencalcium carbonate D3107mg/kg/week. These drugs were applied for14weeks in eachgroups. Collecting blood in one and fourteen weeks, there were measured serummakers of bone metabolism bone alkaline phosphatase (B-ALP) and tartrate-resistantacid phosphatase-5b (TRAP-5b) by ELISA. These rabbits were killed after14weeks,observed the pathological cellular morphology of femurs.Results:1. The basic information: the month of age in each group before treatment wererespectively groupA(5.33±0.20)m, groupB(5.24±0.19)m, groupC (5.23±0.17) m,groupD(5.22±0.23)m、group E(5.31±0.15)m;the weight of each group before treatmentwere respectively groupA (3.53±0.16) kg、 groupB (3.55±0.09)kg、 groupC(3.56±0.14)kg、 groupD (3.52±0.13)kg、 group E (3.51±0.11) kg. There were nosignificant difference between months of age and weight in each group.2. The B-ALP test results: the test results of serum B-ALP and the rate of change were respectively group A35.16±4.19vs35.12±3.12(0.11%); group B35.75±5.27vs26.78±3.07(25.37%); group C35.19±4.32vs30.40±2.22(13.61%); group D35.62±4.68vs30.26±2.37(15.04%); group E34.49±4.63vs28.07±2.62(18.61%). Inthe above groups, except group A, other groups were all reduced after the treatment, butthe decline of group B and E were lower obviously but the difference was notstatistically significant P>0.05.3. The TRAP-5b test results: the test results of serum TRAP-5b and the rate ofchange were respectively group A5.51±1.01vs5.05±0.96(8.34%); group B5.21±1.05vs15.23±3.22(65.79%); group C5.36±1.03vs7.98±1.69(32.83%); group D5.66±1.55vs8.60±1.80(34.18%); group E5.35±1.12vs13.54±1.23(60.48%). In the above groups,except group A, others groups were all increased after the treatment. After treatment thegroups B、 E increased more significantly, compared with before treatment, thedifference was statistically significant P<0.05. Although the groups C and D wereincreased, but the difference was not statistically significant P>0.05.4. The performance of the biopsy: the trabecular bone of group A had continuous,dense, complete, the osteolasts osteoclasts arranged in trabecular bone surface, showingthe form of normal bone tissue. The trabecular bone of group B had fracture, thinningand a large numuber of fat cells fill in the trabecula. The osteoblasts of the trabecularbone surface was not obvious, but visible osteoclasts, showing the faliure pattern ofbone tissue. The trabecular bone of group C had fracture, thinning phenomenon lessthan common, having more active osteoblasts and less osteoclasts. The trabecular boneof group D had rare fracture, thinning phenomenon, having active osteoblasts and lessosteoclasts. The trabecular bone of group E had sparse destruction and a large numuberof fat cells fill in the trabecula. The osteoblasts of the trabecular bone surface is notobvious, but visible osteoclasts. The groups C、D form were between groups A、B. Thegroup E and group B were simiar form.Conclusions: The99Tc-MDP had prevention of rabbit model ofglucocorticoid-induced osteoporosis, its prevention may be achived by promoting theactivity of osteoblasts and inhibit the formation of osteoclasts.
