Experimental Study On Effect Of Xie Shi FA3HF Of Mouse With Advanced Lewis Lung Cancer In P53, Egfr Expression Andshinkage Tumors To Prolong Survival Time Effectively And So On

Objective: Through the establishment of mouse model with advanced Lewis lungcancer, we applicate Xie Shi FA3HF on the model with intervention treatment. Weobserve each mouse with the expression level of P53、EGFR data, the tumor cells withHE staining pattern, tumor weight, tumor shinkage rate, mouse weight and living state,for applicating Xie Shi FA3HF on clinical research better, tamping the experimentalbasis.Methods: We use prepared at concentrations of2.80×107ml-1LLC cellsuspension, with0.1ml transplanted into per C57BL/6J mice by right axillarysubcutaneous, replication of the advanced Lewis lung cancer model, making a total of40. The transplanted mouse are randomly divided into Model group, Xie Shi FA3HFgroup, Bailian Xiaoai Decoction group, Cisplatin group,10mouse in each group.Afterthe tenth day of replication model, Model group: gavage with NS0.9ml, once a day, atotal of10days; Xie Shi FA3HF group: gavage with Xie Shi FA3HF0.9ml, once a day,a total of10days; Bailian Xiaoai Decoction group: gavage with Bailian XiaoaiDecoction0.9ml, once a day, for10days; The Cisplatin group: intraperitoneal injectionwith Cisplatin0.01ml, once a day,5days for a course of treatment, then the next courseof treatment after7days. Finally gavage finished, all mouse are killed. Enucleationmouse’ eyeball for blood, preparation of serum, we use ELISA method to determinatethe levels of P53and EGFR in serum; Removal tumor from each mouse, making ofspecimens and observing the tumor cells with HE staining pattern; Weighting tumor weight and calculating tumor shrinkage rate; Weighting each mouse; Observing eachmouse on living state.Results:1. P53、EGFR datas of Xie Shi FA3HF group、Bailian Xiaoai Decoctiongroup and Cisplatin group are lower than that in Model group, statistical significance (P<0.05). P53、EGFR datas of Xie Shi FA3HF group are lower than that in Bailian XiaoaiDecoction group and Cisplatin group, statistical significance (P <0.05).2. Model groupof tumour cells with HE staining pattern of deterioration degree is higher than that inXie Shi FA3HF group、Bailian Xiaoai Decoction group and Cisplatin group. Xie ShiFA3HF group of tumour cells with HE staining pattern of deterioration degree is lowerthan that in Bailian Xiaoai Decoction group and Cisplatin group.3. Xie Shi FA3HFgroup、Bailian Xiaoai Decoction group and Cisplatin group compared with the Modelgroup, the tumor weight and body weight are both statistical significance (P <0.05).The tumor weight of Xie Shi FA3HF group is less than Bailian Xiaoai Decoctiongroup and Cisplatin group, statistical significance (P <0.05), the tumor shrinkage rateand body weight of Xie Shi FA3HF group are higher than that of Bailian XiaoaiDecoction group and Cisplatin group, statistical significance (P <0.05).Conclusion: Xie Shi FA3HF plays a role in shrinkage tumors on mouse modelwith advanced Lewis lung cancer: containing proliferation and metastasis of tumor cells.Xie Shi FA3HF can enhance mouse’ healthy energy, improve mouse’ quality of life andprolong mouse’ survival time. The inhibition of P53、EGFR data expression levelmaybe is one of the clinical efficacy mechanism of Xie Shi FA3HF.
