Objective:1. Through observe the change of the mice intestinal mucosa form after unblanceand reestablishment of mouse intestinal tract flora, to explore the effect of intestinalflora to intestinal mucosa form.2. Through analyze the changes of mice serum cytokines levels of mouseintestinal flora imbalance and restoration reconstruction, to explore the mechanism ofaction of intestinal bacteria on regulating the immune system.Methods:1. Mice were dealed by intragastic administration of ceftriaxone sodium, accordingto the analysis of cecum flora,8g/kg/d doses,8days to establish dysbacteria model.;2. Utilize10species intestinal predominant flora,such as Bifidobacterium andLactobacillus,dealed by intragastic administration quantitively to re-establish flora;3. Utilize the stool of the normal mice,dealed by intragastic administrationquantitively to re-establish flora;4. Utilize natural recovery to re-establish flora;5. Comparised with normal control group to dysbacteria group〠Flora injectiongroupã€Feces injection groupã€Natural recovery group, morphological changes ofgastrointestinal mucosa were observed by optical microscope and transmission electronmicroscope;6. Comparised with normal control group with dysbacteria groupã€Flora injectiongroupã€Feces injection groupã€Natural recovery group, the changes of secreted IL-1βã€IL-6and TNF-α in serum of each group mice were detected by ELISA. Results:1. Compared with normal control group mouse,after flora imbalance,the mouseintestinal tract mucous membrane morpha was damaged, after three weeks of restorationreconstruction of intestinal flora, compared with natural recovery group the intestinaltract mucous membrane morpha of the flora and feces recharge group mouse have geta better improved,showed a recover trend. The speed of recovery of the flora rechargegroup faster than feces recharge group mouse.2. Compared with normal control group mouse, after flora imbalance, thesecretoryvolume of IL-1βã€IL-6and TNF-αwere higher of mice serum cytokine indysbacteria mice (P<0.05); After three weeks of flora reestablishment, compared withdysbacteria group,the flora and feces recharge group mouse of the secretoryvolumeof IL-1βã€IL-6and TNF-αof mice serum cytokine was step down to turn back(P<0.05),compared with natural recovery group,on two weeks,the flora and feces rechargegroup mouse of the secretoryvolume of IL-1βã€IL-6and TNF-α of mice serumcytokine was step down to turn back(P<0.05).Conclusion:1.Use intestinal the advantage flora recharge and normal mice feces recharge aretwo methods through observation of the intestinal mucosal morphology can be realizedwithin three weeks of recovery fast after severe flora imbalance.2. Use intestinal the advantage flora recharge and normal mice feces recharge aretwo methods are can obvious reduce secretion in the serum cytokine of IL-1βã€IL-6andTNF-α.3. In intestinal mucosal morphology recovery and immune factors regulate thedominant flora recharge group mouse faster than the feces recharge group mouse. |
