| Study on the inhibitory effect of resveratrol on lymphoma cells and Impact of thepro-apoptotic protein caspase-3Objective:Because BJAB cells have the same source with the most of orbital lymphomacells,we choose BJAB cells(a human B cell lymphoma cells)as the research object tostudy the inhibitory effect of resveratrol on BJAB cells and Impact of thepro-apoptotic protein caspase-3.The results of the study can provide an experimentalbasis for Ocular adnexal malt lymphoma(Ocular adnexal mucosa-associated lymphoidtissue lymphoma).Methods:1ã€The BJAB cells are reanimated and cultured.Observe cell morphology andgrowth conditions in a high-powered microscope to understand the characteristics ofthe cells.2ã€The experiment was divided into resveratrol drug groups, doxorubicinpositive control group and blank control group. The concentration of resveratrol druggroups were25μmol/L,50μmol/L,100μmol/L and the concentration ofdoxorubicin positive control group was40μmol/L.Acting on BJAB cells respectivelyto observe morphological changes in24,48,72h. Different groups BJAB cellsproliferation inhibition rate were determined by CCK-8.Observing the proliferationinhibition on BJAB cells in groups of different concentrations of the drug at differenttimes.3ã€Detecting the protein expression of caspase-3by western blot and analysingthe impact of the BJAB cell apoptosis pathway in different concentrations of the drugat different times.Results:1ã€BJAB cells grew well and passed stably.The shape of cells were round orellipse and the caryons were round.2〠Different drug concentrations of resveratrol drug groups and doxorubicinpositive control group aiming at BJAB cells24h,48h,72h,can inhibit cell proliferationin varying degrees.CCK-8assay OD values were lower than the control group, andstatistical analysis, the gap was statistically significant (P <0.05).3ã€Western blot test results shows that, compared with the blank control group,different drug concentrations of resveratrol drug groups and doxorubicin positivecontrol group aiming at BJAB cells24h,48h,72h,caspase-3protein expression risingin varying degrees.Conclusion:1ã€Resveratrol within a certain concentration range can inhibit BJAB cellproliferation and different concentrations of resveratrol effect different at differenttimes. Concentration and time can affect cell proliferation, in which the time-dependent effectis obvious.When the concentrationof resveratrol drug group were100μmol/L,showing time-dependent.Although time-dependent were not exsit when the concentration ofresveratrol drug group were25μmol/L and50μmol/L,however,after72h,theconcentration of50μmol/L lower cell activity and cell death more, so theconcentration will affect cell activity, affect cell proliferation.2ã€Resveratrol within a certain concentration can inhibit cell proliferation byaffecting the expression of caspase-3protein at different times.The rising of caspase-3protein expression on different drug concentrations of resveratrol drug groups werenot obvious,but at72h, caspase-3protein expression was significantly elevated in adose-dependent.3ã€According to the results of this experiment, we concluded that resveratrol alsohas a certain role in the inhibition of cell proliferatio for Ocular adnexal maltlymphoma.Then the results can supply the experimental and theoretical basis for theexperimental study in the future. |
