Studies On Apoptosis Of Human T Lymphoma Jurkat Cell Line Induced By Resveratrol

Aim:To establish an apoptotic model induced by resveratrol(Res) in the T lymphoma Jurkat cell line in order to describe the characteristic morphological alteration of apoptosis ,such as chromatin margination or condensation, nuclear fragmentation and apoptotic bodies and so on ; To examine and analyze the alteration of apoptosis treated with resveratrol at various concentrations and different time;and to explore the mechanism of resveratrol -induced apoptosis. Methods:Firstly,analyzing the DNA fragments by agarose gel electrophoresis to testify apoptosis and the time of apoptosis. Cells were observed under fluorescence ,electron transmission microscopy,and observed after Wright-Giemsa dyeing. Secondly, MTT assay method were used to identify the alteration of apoptosis treated with resveratrol at various concentrations and different time;the flow cytometry (FCM) was used to investigate the apoptotic rate , changes of cell cycle distribution and the alteration of apoptosis treated with resveratrol at various concentrations.Lastly, using the West blotting method to examine the changes of β -actin,bax,caspase-3,bcl-2,Bcl-xl and AIF in the T lymphoma Jurkat cell during the apoptosis induced by resveratrol. Results and conclusion:Res can cause T lymphoma Jurkat cell line to undergo apoptosis in concentration and time dependent manner. The characteristic morphological alteration of apoptosis can be observed such as chromatin margination , chromatin condensation, nuclear fragmentation,the vacuole cytoplasm and apoptotic bodies. Res can cause nearly all T lymphoma Jurkat cells undergo the latter part of apoptosis after 48 hours. 0.2 mmol/L Res can cause 1/3 T lymphoma Jurkat cells undergo apoptosis after 24 hours. It indicates that the growth of T lymphoma Jurkat cells was blocked at G0/G1 phase during the cell cycle.It's function may be attributed to the decreased expression of bcl-xl in the cells ,so that it cannot form the dimer to hold back the apoptosis.Therefore,the Bax and AIF can incite the next signal molecule.At last, the cells begin to apoptosis on caspase-3 depended type.
