Objective To investigate the effect of48%chlorpyrifos on cholinesterase activity, signal transduction pathway in rats.Methods Rats were randomly divided into low-dose,middle-dose, high-dose(groups and saline control group. The rats were treated by oral gavage for28days. The activity of whole blood cholinesterase were detected with ferric chloride colorimetric method, microscopic observe the ill brain tissue of the rats, the expression of the receptor of advanced glycation end-products (RAGE) and protein kinase C(PKC) were detected with the immunohistochemistry in brain tissue, the expression of Gq protein were detected with the Western blot in brain tissue.Results The activity of whole blood cholinesterase degrades with increased exposure, the activity of middle and high-dose group was statistically significantcompared with the control group. The results of brain tissue of the rats microscopic observation:inflammation of lymph cell concentrate. The expression of RAGE, Gq protein, PKC increases with increased exposure.Conclusions Chlorpyrifos injuries the body by inhibiting the activity of cholinesterase. The high expression of RAGE, Gq, PKC may be involved nervous system pathological signal transduction in chlorpyrifos poisoning.
